Chemokines control the migratory patterns and positioning of immune cells to organize immune responses to pathogens. However, many chemokines have been associated with systemic autoimmune diseases that have chronic IFN signatures. We report that a series of chemokines, including CXCL4, CXCL10, CXCL12, and CCL5, can superinduce type I IFN (IFN-I) by TLR9-activated plasmacytoid DCs (pDCs), independently of their respective known chemokine receptors. Mechanistically, we show that chemokines such as CXCL4 mediate transcriptional and epigenetic changes in pDCs, mostly targeted to the IFN-I pathways. We describe that chemokines physically interact with DNA to form nanoparticles that promote clathrin-mediated cellular uptake and delivery of DNA in the early endosomes of pDCs. Using two separate mouse models of skin inflammation, we observed the presence of CXCL4 associated with DNA in vivo. These data reveal a noncanonical role for chemokines to serve as nucleic acid delivery vectors to modulate TLR signaling, with implications for the chronic presence of IFN-I by pDCs in autoimmune diseases.

The performance of nanomaterials is significantly determined by the interfacial microenvironment, in which a surfactant plays an essential role as the adsorbent, but its involvement in the interfacial reaction is often overlooked. Here, it was discovered that citrate and ascorbic acid, the two primarily used surfactants for colloidal gold nanoparticles (Au NPs), can spontaneously undergo catalytic reaction with trace-level nitrogenous residue under ambient environment to form oxime, which is subsequently cleaved to generate CN or a compound containing the -CN group. Such a catalytic reaction shows wide universality in both reactants, including various carbonaceous and nitrogenous sources, and metal catalysts, including Au, Ag, Fe, Cu, Ni, Pt, and Pd NPs.

In this work, we synthesize a quinoline-based heptamethine cyanine, QuCy7, with sulfonate groups to enhance water solubility. This dye demonstrates exceptional near-infrared absorption beyond 750 nm, accompanied by photothermal properties but low photostability. Encapsulating QyCy7 with polyethylene glycol to form nanopolymer, QuCy7@mPEG NPs, addresses the issue of its photoinstability.

Herein, porous SnO microspheres in a three-dimensional (3D) hierarchical architecture were successfully synthesized via a facile hydrothermal route utilizing d-(+)-glucose and cetyltrimethylammonium bromide (CTAB), which act as reducing and structure-directing agents, respectively. Controlled adjustment of the CTAB to glucose mole ratio, reaction temperature, reaction time, and the calcination parameters all provided important clues toward optimizing the final morphologies of SnO with exceptional structural stability and reasonable monodispersity. Electron microscopy analysis revealed that microspheres formed were hierarchical self-assemblies of numerous primary SnO nanoparticles of ∼3-8 nm that coalesce together to form nearly monodispersed and ordered spherical structures of sizes in the range of 230-250 nm and are appreciably porous.
