Respiratory manifestations of sickle cell disease in children: a comprehensive review for the pediatrician.

Expert Rev Respir Med

January 2025

Department of Surgery, Dentistry, Pediatrics and Gynaecology, Pediatric Division, University of Verona, Verona, Italy.

Introduction: Sickle cell disease (SCD) is an inherited hemoglobinopathy characterized by the production of sickle hemoglobin, leading to red blood cells sickling and hemolysis in hypoxic conditions. The resulting acute and chronic endothelial inflammation leads to chronic organ damage. Respiratory manifestations in SCD usually start from childhood and represent the leading causes of morbidity and mortality.

Article Synopsis
  • The pathogenesis of long COVID (LC) involves uncertainty, complicating the search for effective therapies.
  • The hypothesis suggests that chronic damage to the body's anti-inflammatory mechanisms, particularly through the vagus nerve, HPA axis, and mitochondrial function, plays a crucial role in LC development.
  • The theory posits that SARS-CoV-2 alters these systems at various levels, leading to persistent inflammation due to impaired anti-inflammatory responses from acetylcholine and cortisol, warranting further investigation into glucocorticoid receptor sensitivity and potential long-term epigenetic effects.

Skeletal muscle adaptations and post-exertional malaise in long COVID.

Trends Endocrinol Metab

December 2024

Department of Human Movement Sciences, Faculty of Behavioural and Movement Sciences, Vrije Universiteit Amsterdam, The Netherlands; Amsterdam Movement Sciences Research Institute, Amsterdam, The Netherlands. Electronic address:

When acute SARS-CoV-2 infections cause symptoms that persist longer than 3 months, this condition is termed long COVID. Symptoms experienced by patients often include myalgia, fatigue, brain fog, cognitive impairments, and post-exertional malaise (PEM), which is the worsening of symptoms following mental or physical exertion. There is little consensus on the pathophysiology of exercise-induced PEM and skeletal-muscle-related symptoms.

Endothelial Dysfunction and Impaired Wound Healing Following Radiation Combined Skin Wound Injury.

Int J Mol Sci

November 2024

Radiation Combined Injury Program, Armed Forces Radiobiology Research Institute, Uniformed Services University of the Health Sciences, Bethesda, MD 20814, USA.

Article Synopsis
  • There are currently no FDA-approved treatments for radiation combined injury (RCI) due to a limited understanding of its mechanisms.
  • Research indicates that endothelial dysfunction contributes to higher mortality and delayed healing in RCI, as evidenced by increased body weight loss and skin wound complications in RCI mice compared to those with only skin wounds or radiation injury alone.
  • The study also suggests that targeting endothelial dysfunction and the TGF-β signaling pathway could offer potential therapeutic strategies to improve healing and outcomes in patients with RCI.
