Understanding cellular responses to genetic perturbations is essential for understanding gene regulation and phenotype formation. While high-throughput single-cell RNA-sequencing has facilitated detailed profiling of heterogeneous transcriptional responses to perturbations at the single-cell level, there remains a pressing need for computational models that can decode the mechanisms driving these responses and accurately predict outcomes to prioritize target genes for experimental design. Here, we present scLAMBDA, a deep generative learning framework designed to model and predict single-cell transcriptional responses to genetic perturbations, including single-gene and combinatorial multi-gene perturbations.

Unlabelled: Since the advent of smartphones, peer interactions over digital platforms have become a primary mode of socializing among adolescents. Despite the rapid rise in digital social activity, it remains unclear how this dramatic shift has impacted adolescent social and emotional experiences. In an intensive, longitudinal design ( = 26, = 206 monthly observations for up to 12 months, 12-17 years), we used digital phenotyping methods to objectively measure within-person fluctuations in smartphone use (screen time, pickups, notifications) across different categories (social media, communication, entertainment, games) and examined their prospective, bidirectional associations with positive and negative mood.

In agroecosystems, nitrous oxide (N₂O) emissions are influenced by both microbiome composition and soil properties, yet the relative importance of these factors in determining differential N₂O emissions remains unclear. This study investigates the impacts of these factors on N₂O emissions using two primary agricultural soils from northern China: fluvo-aquic soil (FS) from the North China Plain and black soil (BS) from Northeast China, which exhibit significant differences in physicochemical properties. In non-sterilized controls (NSC), we observed distinct denitrifying bacterial phenotypes between FS and BS, with BS exhibiting significantly higher N₂O emissions.
