Microridges are laterally elongated actin-based protrusions arranged in striking maze-like patterns on the apical surfaces of mucosal epithelial cells. Recent studies have begun to reveal the molecular and mechanical factors that regulate microridge morphogenesis and allow them to adopt their unique properties. Microridges form from the coalescence of short actin-filled precursor protrusions called pegs. Microridge morphogenesis requires the establishment of apicobasal polarity, cortical myosin contraction, and Arp2/3 activity. Mature microridges contain branched actin networks, keratin filaments, and plakin cytolinkers that likely connect the two cytoskeletal elements. Once formed, microridges rearrange by fission and fusion to form increasingly regular patterns. Their highly organized arrangement provides an exciting system in which to study the interplay between molecular signaling and physical forces in the formation of subcellular patterns.

Actin filaments and microtubules create diverse cellular protrusions, but intermediate filaments, the strongest and most stable cytoskeletal elements, are not known to directly participate in the formation of protrusions. Here we show that keratin intermediate filaments directly regulate the morphogenesis of microridges, elongated protrusions arranged in elaborate maze-like patterns on the surface of mucosal epithelial cells. We found that microridges on zebrafish skin cells contained both actin and keratin filaments.

This study was conducted with the tongue samples of different life stages of hilsa, that is, adult Marine hilsa, adult Riverine hilsa, and Riverine juvenile hilsa, respectively. Three types of taste buds (Types I, II, and III based on their elevation from the epithelium at different levels) of the tongue, which may be to ensure full utilization of the gustatory ability of the fish were rocorded. Presence of specific taste buds indicate that the fish hilsa dwells in turbid waters with a possible preference toward diatom like planktonic food source.
