An efficient route to -alkylated 3,4-dihydroisoquinolinones with substituents at the 3-position.

A facile synthetic procedure for the production of -alkylated 3,4-dihydroisoquinolinone derivatives is described. The desired products were obtained by -alkylation of 3,3'-dimethyl-3,4-dihydroisoquinoline derivatives followed by oxidation of the resulting iminium salts. Reaction conditions for both steps were very mild and the desired cyclization products could be obtained in good yield. This strategy allows the generation of -substituted 3,4-dihydroisoquinolinone derivatives with substituents at the 3-position.

SARS-CoV-2 3CL protease (Main protease) and human cathepsin L are proteases that play unique roles in the infection of human cells by SARS-CoV-2, the causative agent of COVID-19. Both proteases recognize leucine and other hydrophobic amino acids at the P position of a peptidomimetic inhibitor. At the P position, cathepsin L accepts many amino acid side chains, with a partial preference for phenylalanine, while 3CL-PR protease has a stringent specificity for glutamine or glutamine analogues.

Discerning and quantifying the critical biothiols cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) are vital for understanding their synergistic roles in biological systems. In this study, we synthesized a series of phenylethynylcoumarin fluorescent probes with varied structures to investigate the mechanisms underlying biothiol detection. We found that different substituents (-OCH, -H, -CN) at the -position of the phenylacetylene, combined with an aldehyde group at the 3-position of the coumarin, significantly affected the probes' reactivity and produced distinct response patterns toward biothiols.

Various new tris(pyrazolyl)alkanes of the class R'CTp (R' = Me, Et, Pr, iBu; R = Et, Pr, Cy, BuPh, Ph; Pr = cyclopropyl, Cy = cyclohexyl, BuPh = -butylphenyl) were synthesised and their reactivity towards Mg(AlMe) was examined. Along with new examples of recurring structural motifs, such as separated ion pairs and "metal in a box" complexes, , [(MeCTp)Mg][AlMe], several magnesium complexes with new structural features/compositions were obtained. Treatment of the "metal in a box" species [(MeCTp)Mg][AlMe] with THF donor gave the terminal methyl complex [(MeCTp)MgMe(thf)][AlMe].

We describe herein a facile method to access 2,3-epoxyesters with fluorine-containing substituents at their 3-position starting from the corresponding enoates by utilization of the low-costed and easy-to-handle reagent, NaOCl·5HO. Because very little has been disclosed about the reactivity of such 2,3-epoxyesters, their epoxy ring opening by a variety of nucleophiles was carried out and we succeeded in clarifying these chemo- as well as regioselective processes proceeding via the S2 mechanism to mainly afford 2-substituted 3-hydroxyesters usually in a highly selective manner.
