The present study aimed to develop and validate an advanced image stitching algorithm integrated with chemical imaging at the nanometre scale. This was applied to track the swelling, erosion, drug release and changes in surface texture of a swelling-controlled release system. The technique involves the delivery and withdrawal of a liquid droplet from the surface of the tablet alongside capturing multiple images of tablet surface using white light profilometry. The recovered liquid was then subject to chemical analysis for the quantification of drug and HPMC. The multiple images acquired during drug release were stitched together using an algorithm developed to generate a full tablet surface. New methods for swelling analysis (regional point, area and multiple regional analysis techniques) were also successfully developed. The results exhibited the exceptional capability of this technique for providing quantitative information regarding swelling, erosion, drug release and surface topography, hence negating the need for separate investigations. Moreover, it can also be anticipated that this technique may have potential use in other fields where surface dissolution, erosion and swelling have significant impact.

A low oxygen level in solid tumors is behind the modern concept of selective chemotherapy by hypoxia-activated prodrugs, such as heteroleptic complexes of transition metals (cobalt(III), iron(III) or platinum(IV)) with bi- or tetradentate ligands and an anticancer drug molecule as a co-ligand. A series of new cobalt(III) complexes [Co(LR)(esc)]ClO with esculetin (6,7-dihydroxycoumarin) and 2,2'-bipyridines (2,2'-bipy) functionalized by different substituents R were probed in the hypoxia-activated delivery of this model anticancer drug. Their combined study by cyclic voltammetry and NMR spectroscopy allowed identifying linear correlations of the electrochemical reduction potentials and the rate of the hypoxia-activated dissociation of [Co(LR)(esc)]ClO with the Hammett constants of the substituents in 2,2'-bipy ligands.

Gold Nanorods Decorated by Conjugated Microporous Polymers for Infrared Responsive Cytostatic Drug Delivery.

Near-infrared (NIR) controlled drug delivery systems have drawn a lot of attention throughout the past few decades due to the deep penetration depth and comparatively minor side effects of the stimulus. In this study, we introduce an innovative approach for gastric cancer treatment by combining photothermal infrared-sensitive gold nanorods (AuNRs) with a conjugated microporous polymer (CMP) to create a drug delivery system tailored for transporting the cytostatic drug 5-fluorouracil (5-FU). CMPs are fully conjugated networks with high internal surface areas that can be precisely tailored to the adsorption and transport of active compounds through the right choice of chemical functionalities.
