DNA replication profiling by molecular combing on single DNA fibers.

STAR Protoc

Developmental Therapeutics Branch, Center for Cancer Research, NCI, NIH, Bethesda, MD, USA.

Published: June 2022

In this protocol, the progression of DNA synthesis is profiled at a single-molecule resolution. DNA fibers are uniformly stretched on silanized coverslips, and replicating DNA can be traced with thymidine analogs using specific antibodies against distinct analogs. Single DNA fibers are visualized by an anti-single stranded DNA antibody. The protocol can be used to study DNA replication dynamics, the cellular response to replication stress, and replication fork progression at specific chromosomal regions when combined with fluorescent hybridization. For complete details on the use and execution of this protocol, please refer to Conti et al. (2007), Fu et al. (2021), Kaykov et al. (2016), Redmond et al. (2018), and Schwob et al. (2009).

Objective: We assessed the impact of a food-provisioning intervention on diet quality in children with obesity.

Methods: Participants (n = 33, aged 6-11 years) were randomly assigned to either usual care (intensive health behavior and lifestyle treatment) or intervention (usual care + food provisioning; high-fiber, low-dairy diet) for 4 weeks. The primary outcome was a change in child diet quality at Week 4.

Transcription factors and genome biases in polyploid crops.

Adv Protein Chem Struct Biol

January 2025

National Agri-Food Biotechnology Institute, Knowledge City, Mohali, Punjab, India. Electronic address:

Nuclear protein transcription factors (TFs) regulate all biological processes in plants and are necessary for gene regulation. The transcription of genes during plant growth and development and their response to environmental cues are regulated by TF binding to specific promoter regions in the genomic DNA. Polyploid plants with several sets of chromosomes frequently display intricate genomic biases concerning TF expression.

An erbium-doped fiber ring laser based on a single-mode fiber-no-core fiber-single-mode fiber (SMF-NCF-SMF) structure was constructed and experimentally demonstrated for label-free DNA hybridization measurement. The SMF-NCF-SMF structure acts as a sensing element and a filter to select the laser wavelength. The proposed fiber ring laser sensor exhibits a high optical signal-to-noise ratio (SNR, >50 dB) and narrow full width at half maximum (FWHM, <0.

Atrial remodeling is a major pathophysiological mechanism of atrial fibrillation (AF). Atrial remodeling progresses with aging and background diseases, including hypertension, heart failure, and AF itself. However, its mechanism of action and reversibility have not been completely elucidated.

DNA nanostructures (DNs) have gained popularity in various biomedical applications due to their unique properties, including structural programmability, ease of synthesis and functionalization, and low cytotoxicity. Effective utilization of DNs in biomedical applications requires a fundamental understanding of their interactions with living cells and the mechanics of cellular uptake. Current knowledge primarily focuses on how the physicochemical properties of DNs, such as mass, shape, size, and surface functionalization, affect uptake efficacy.
