Septin filaments assemble into high-order molecular structures that associate with membranes, acting as diffusion barriers and scaffold proteins crucial for many cellular processes. How septin filaments organize in such structures is still not understood. Here, we used fission yeast to explore septin filament organization during cell division and its cell cycle regulation. Live-imaging and polarization microscopy analysis uncovered that septin filaments are initially recruited as a diffuse meshwork surrounding the acto-myosin contractile ring (CR) in anaphase, which undergoes compaction into two rings when CR constriction is initiated. We found that the anillin-like protein Mid2 is necessary to promote this compaction step, possibly acting as a bundler for septin filaments. Moreover, Mid2-driven septin compaction requires inputs from the septation initiation network as well as CR constriction and the β(1,3)-glucan synthase Bgs1. This work highlights that anillin-mediated septin ring assembly is under strict cell cycle control.

Septin 9 (SEPT9), a GTPase, known as the fourth cytoskeleton, is widely expressed in various cells and tissues. The functions of SEPT9 are partly similar to other cytoskeletons as a structure protein. Further, SEPT9 can interact with other cytoskeletons, participating in actin dynamics and microtubule regulation.

Article Synopsis
  • Ocular hypertension, a key risk factor for glaucoma, is linked to increased activity and stiffness in the trabecular meshwork (TM) due to cellular interactions.
  • Previous research identified high levels of septin-9 in TM cells under hypertensive conditions, prompting studies on its overexpression, deficiency, and drug targeting effects.
  • Findings indicate that septin-9 impacts intraocular pressure (IOP) regulation, suggesting that modulating septin cytoskeletal organization could be a potential new treatment strategy for glaucoma.

Colletotrichum gloeosporioides is a model plant pathogenic fungus, and the appressoria are the main infection structures integral to the pathogenic process. Septin proteins play fundamental roles in facilitating shape alteration and organizing the F-actin cytoskeleton, thereby aiding the invasive growth of various fungi. Herein, we examined the roles of four septin-coding genes (CgSEP3, CgSEP4, CgSEP5, and CgSEP6) in C.

Septins are involved in many important cellular processes, and septin dysfunction has been implicated in various pathologies, such as cancer. Like other components of the cytoskeleton -F-actin, microtubules, and intermediate filaments-septins can self-assemble into filaments and higher-order structures. These non-polar filaments are assembled from complex and variable multimeric building blocks.
