Candida albicans (C. albicans) is a fungal pathogen that causes infections of the wet body surfaces and the blood in immunocompromised patients or individuals with imbalanced microflora. Since the cases of clinically meaningful candidosis are on the rise, efficient С. albicans therapy is in a high demand. Informed drug design requires well-characterized С. albicans targets, including these aimed at disrupting its post-translational modifications. C. albicans ORF19.2286 gene encodes an ortholog of human deoxyhypusine hydroxylase (DOHH). Here, this ORF was cloned from the SC5314 strain and re-expressed in Escherichia coli as sGB1 - CaDOHH construct with 6xHis tag on the N-terminus of the fusion protein, then purified, and GB1-tag was removed with Tobacco etch virus (TEV) protease. Several amino acid sequence differences between C. albicans and animal DOHHs were noted, and are useful for a selection of the binding sites for antimicrobials in CaDOHH. We present the protocol for the heterologous expression and purification of C. albicans DOHH, which is suitable for further crystallization.

Lung metastases occur in up to 54% of patients with metastatic tumours. Contributing factors to this high frequency include the physical properties of the pulmonary system and a less oxidative environment that may favour the survival of cancer cells. Moreover, secreted factors from primary tumours alter immune cells and the extracellular matrix of the lung, creating a permissive pre-metastatic environment primed for the arriving cancer cells.

The unique amino acid hypusine [N-(4-amino-2-hydroxybutyl)lysine] is exclusively formed on the translational regulator eukaryotic initiation factor 5A (eIF5A) via a process coined hypusination. Hypusination is mediated by two enzymes, deoxyhypusine synthase (DHPS) and deoxyhypusine hydroxylase (DOHH), and hypusinated eIF5A (eIF5A) promotes translation elongation by alleviating ribosome pauses at amino acid motifs that cause structural constraints, and it also facilitates translation initiation and termination. Accordingly, eIF5A has diverse biological functions that rely on translational control of its targets.

This review highlights recent progress in the application of biotin probes of homoisoflavonoids for identifying binding proteins and elucidating mechanisms of action. Notably, homoisoflavonoids exhibit antiangiogenic, anti-inflammatory, and antidiabetic effects.
