T cells play essential roles in antitumor therapy. Via gene engineering technique to enhance tumor-antigen specificity, patient peripheral blood-derived T cells (PBT) show encouraging clinical outcomes in treating certain blood malignancies. However, the high costs, functionality exhaustion, and disease-condition-dependent availability of PBT prompt the attempts of exploring alternative T cell sources. Theoretically, induced T cells from pluripotent stem cells (PSC) are ideal candidates that integrate plenty of advantages that primary T cells lack, including unlimited off-the-shelf cell source and precision gene editing feasibility. However, researchers are still struggling with developing a straightforward protocol to induce functional and immunocompetent human T cells from PSC. Based on stromal cell-expressing or biomaterial-presenting Notch ligands DLL1 or DLL4, natural and induced blood progenitors can differentiate further toward T lineage commitment. However, none of the reported T induction protocols has yet translated into any clinical application, signaling the existence of numerous technical barriers for regenerating T cells functionally matching their natural PBT counterparts. Alternatively, new approaches have been developed to repopulate induced T lymphopoiesis via reprogramming or transplanting induced T cell precursors. Here, we review the most recent progress in the T cell regeneration field, and the remaining challenges dragging their clinical applications.

Haploinsufficiency for is associated with congenital heart disease (CHD) with variable comorbidity of pancreatic or diaphragm defects, although the etiology of disease is not well understood. Here, we used cardiac directed differentiation from human embryonic stem cells (hESCs) as a platform to study GATA6 function during early cardiogenesis. GATA6 loss-of-function hESCs had a profound impairment in cardiac progenitor cell (CPC) specification and cardiomyocyte (CM) generation due to early defects during the mesendoderm and lateral mesoderm patterning stages.

Inspired by the continuous regeneration of enzymes in living organisms that enables sustained long-term enzymatic reactions, we propose an active site regeneration strategy to rapidly and in-situ recover the catalytic activity of oxygen evolution reaction (OER) for the electrode through switching anode solution, to substantially prolong the service life of the electrolytic cell. The OER activity of anodic NiFe(S)/NM catalyst synthesized by coating S-doped NiFe-LDH on nickel mesh (NM) exhibited excellent initial activity, then experienced gradual active site and activity losses due to the nanosheet detachment from NMs after a long-term OER process. Attractively, such a damaged catalyst and its active sites can be effectively regenerated by in-situ redox reaction followed by electrochemical oxidation through switching the anode into the different solutions, resulting in the full recovery of the OER activity to its initial value.

We describe the novel use of dynamic sentinel node biopsy (DSNB) in five patients with primary urethral squamous cell carcinoma (U-SCC) and no evidence of inguinal node disease across two centers in North America and Europe between 03/2021 and 06/2024. Each of these referral centers sees over 75 cases of penile cancer per year and approximately 10 cases of U-SCC per year. Patients underwent DSNB concomitant to surgical resection of the primary tumor (n=3), or in a deferred manner (n=2), six weeks after primary surgery.

The primary clinical challenges associated with postoperative maxillofacial osteosarcoma include high mortality rates and significant local recurrence. Additionally, patients often exhibit substantial bone defects that are incapable of self-healing, necessitating the implantation of scaffolds. Multifunctional hydrogels, which enable sustained local release of therapeutic agents and enhance scaffold surface properties, demonstrate significant potential for the postoperative management of maxillofacial osteosarcoma.
