AI Article Synopsis

  • A new flavanone-flavonol dimer named gnidiflavanone-flavonol and 10 other known compounds were isolated from the roots of a plant in the Thymelaeceae family.
  • The structures of these compounds were determined using advanced methods like NMR and mass spectrometry, with some compounds being reported for the first time from this plant family.
  • In terms of biological activity, the crude extract and isolated compounds were tested for antimicrobial and antiplasmodial effects, with two compounds showing promising antiplasmodial activity against both chloroquine-sensitive and resistant strains.

Article Abstract

A new 3,8''-flavanone-flavonol dimer gnidiflavanone-flavonol () and 10 known compounds (-), including four rare primula-type flavones -, were isolated from the roots of . Compounds - and were reported for the first time from the plant family Thymelaeceae. Structures of the isolated compounds were established by spectroscopic data, including 1D and 2D NMR (COSY, HMBC, HSQC and ROESY) and mass spectrometry, as well as by the comparison with literature data. The crude roots extract and isolated compounds were evaluated for antimicrobial and antiplasmodial activities. Among isolated compounds, 6-hydroxyflavone () and 6--acetylflavone () showed antiplasmodial activity against chloroquine-sensitive (D6) and chloroquine-resistant (W2) strains of .

Introduction: The developed domestic retrodipeptide analogue of cholecystokinin tetrapeptide (CCK) (N-(6-phenylhexanoyl)-glycyltryptophan amide, or compound GB-115) with antagonistic properties in relation to CCK1 receptors has anxiolytic activity previously shown in preclinical and clinical studies. The aim of the study was to evaluate the anxiolytic effect of GB-115 as a tablet form with subchronic oral administration in comparison with phenazepam in nonhuman primates.

Materials And Methods: The study was conducted on four male rhesus monkeys (Macaca mulatta) aged 5.

Five previously unreported 5-hydroxymethylfurfural (5-HMF) derivatives, including chinenfurfurals A () and B () as 5-HMF-citric acid hybrids and chinenfurfurals C-E (-) as 5-HMF oligomers, as well as four known analogues (-), were isolated from the fruits of a well-known Chinese herbal species . The structures of these furfural compounds were established by detailed analyses of spectroscopic data especially HRMS and NMR, and it is the first report of furfural type constituents from the title species. The anti-inflammatory property of them was further evaluated by testing their inhibition against the production of nitric oxide in lipopolysaccharide-activated murine RAW264.

Miniaturized mass spectrometers offer significant potential for in situ analysis due to their high specificity and portability. In traditional data-dependent acquisition (DDA) mode, precursor ions for tandem analysis are selected based on the full-scan mass spectrum. However, in situ applications often require the direct analysis of complex samples without extensive sample pretreatment, making them susceptible to chemical noise that can result in false negatives.

Monomer compounds from natural products are the major source of active pharmaceutical molecules, which provide great opportunities for discovering of new drugs. However, natural products contain a large number of rather complex compounds. It is difficult to obtain high-purity monomer compounds from complex natural products.

A comprehensive strategy, including spectroscopic, molecular simulation, proteomics, and bioinformatics techniques, was employed to investigate a novel triazole, 5-(4-methoxyphenyl)-1-phenyl-1H-1,2,3-triazole, its interactions with high-abundance blood proteins, and identification of low-abundance proteins. The binding constants and thermodynamic parameters of the triazole to two high-abundance blood globular proteins, human serum albumin, and human immunoglobulin G (HIgG), were obtained by spectroscopic techniques and computational chemistry. The two-dimensional gel electrophoresis in combination with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry was employed to isolate and identify differentially expressed low-abundance proteins in human blood serum samples following exposure to the triazole.
