Rheumatoid arthritis (RA) is a chronic inflammatory disease that mainly targets the synovial membrane, thus causing stiffness, deformity and dysfunction of joints. To date, no effective anti‑inflammatory treatments are available for RA. Piceatannol (PIC) is a natural derivative of resveratrol, which has been reported to attenuate the inflammatory response. To evaluate the effect of PIC on RA and to determine the underlying molecular target of PIC, both and experiments were performed in the present study. A CIA rat model was established to evaluate the therapeutic effects of PIC. TNF‑α, IL‑1β and IL‑6 levels in blood were measured by ELISA. Western blotting, immunofluorescence analysis and reverse transcription‑quantitative PCR (RT‑qPCR) were used to analyze the expression levels of protein and mRNA. , RA‑fibroblast‑like synoviocytes (FLSs) were pretreated with PIC and subsequently stimulated with TNF‑α. The results revealed that PIC significantly upregulated the expression levels of proapoptotic proteins such as Bax and cleaved caspase‑3. PIC also significantly reduced the production of proinflammatory cytokines, including PGE2, IL‑6 and IL‑1β, and significantly downregulated the expression of cyclooxygenase‑2 at both the mRNA and protein expression levels. Furthermore, PIC downregulated the expression of MMP‑3 and MMP‑13, which have been found to be highly expressed in the synovium of patients with RA. Mechanistically, PIC was capable of significantly downregulating the expression levels of proteins involved in the NF‑κB and MAPK signaling pathways. The results of the experiments using a rat collagen‑induced arthritis model demonstrated that PIC decreased the arthritis score and exerted beneficial effects in cartilage and significantly reduced the expression of MMP‑13. In conclusion, the findings of the present study revealed that PIC could suppress the inflammatory response, promote apoptosis, and exert a significant regulatory effect on the NF‑κB and MAPK signaling pathways in RA‑FLSs. Therefore, PIC may represent a potential drug for the future treatment of RA.

Purpose: To investigate the therapeutic efficacy of BEZ235, a dual PI3K/mTOR inhibitor, in suppressing pathological neovascularization in an oxygen-induced retinopathy (OIR) mouse model and explore the role of cyclin D1 in endothelial cell cycle regulation.

Methods: Single-cell RNA sequencing was performed to analyze gene expression and cell-cycle alterations in retinal endothelial cells under normoxic and OIR conditions. The effects of BEZ235 on human umbilical vein endothelial cells (HUVECs) and human retinal microvascular endothelial cells (HRMECs) were evaluated by assessing cell viability, cell-cycle progression, proliferation, migration, and tube formation.

Endometrial cancer (EC) is the second most frequent gynecological malignancy and the sixth most common women's cancer worldwide. EC incidence rate is increasing rapidly. Apart from the classical, we should consider angiogenesis and hypoxia-related genes as a reason for EC manifestation and progression.

Non-alcoholic fatty liver disease (NAFLD) is a disease with various levels varying from fatty liver steatosis to acute steatosis which is non-alcoholic steatohepatitis (NASH), which can develop into hepatic failure, as well as in some conditions it can develop into hepatocellular carcinoma (HCC). In the NAFLD and NASH context, aberrant microRNA (miRNA) expression has a thorough contribution to the incidence and development of these liver disorders by influencing key biological actions, involving lipid metabolism, inflammation, and fibrosis. Dysregulated miRNAs can disrupt the balance between lipid accumulation and clearance, exacerbate inflammatory responses, and promote fibrogenesis, thus advancing the severeness of the disorder from simple steatosis to more complex NASH.

The use of cell lines as alternative models for environmental physiology studies opens a new window of possibilities and is becoming an increasingly used tool in marine research to fulfil the 3R's rule. In this study, an embryonic monoclonal stem cell line obtained from a marine teleost (gilthead seabream, Sparus aurata) was employed to assess the effects of photoperiod (light/dark cycles vs constant dark) and light spectrum (white, blue, green, blue/green and red lights) on gene expression and rhythms of cellular markers of proliferation, DNA repair, apoptosis and cellular/oxidative stress by RT-qPCR and cosinor analyses. The results obtained revealed the optimal performance of cells under blue light (LDB), with all the genes analysed showing their highest RNA expression levels and most robust daily variations/rhythms in this condition.

Hepatocellular carcinoma (HCC) is a primary liver malignancy characterized by high morbidity and mortality. Recently, ferroptosis has been recognized as an important factor in regulating cell growth in HCC. However, the role of ferroptosis-related genes in HCC remains unclear.
