In this contribution, the analytical potential of total reflection X-ray fluorescence (TXRF) instrumentation has been evaluated for the determination of major and trace elements in milk powder. TXRF allows the possibility of direct analysis of solid suspensions without the need for a digestion process and therefore it can be a potential analytical candidate for simple and cost-effective analysis. A detailed study to select sample preparation and measurements conditions was carried out. Different quantification approaches (including internal standardization and empirical calibration) were also tested. Finally, the developed TXRF methods (W anode) were validated by a strict comparison with the data from the reference methods on a set of twenty-three samples using robust statistics. Results showed that acceptable results can be obtained for K, Ca, Fe and Zn determination if using adequate calibration approaches. Otherwise, only screening results can be obtained for light elements (P and Cl) in milk powder samples.

Extracellular vesicles (EVs) are promising therapeutic carriers for their ideal nano-size and intrinsic biocompatibility, while rapid clearance and limited targeting ability are the major setbacks of EVs. With minimal absorption into the systemic circulation, inhalation for pulmonary disease therapy minimizes off-target toxicity to other organs and offers a safe and effective treatment for respiratory disorders. This study integrates proteome analysis of human breast milk (HBM) from a homogeneous group of mothers who are of similar age and live in the same geographical area, along with an analysis of essential and potentially toxic elements in HBM in relation to lifestyle and environmental factors. This preliminary proteomic study, which examined 11 samples of HBM from lactating women, identified a total of 1619 proteins across all samples, revealing significant differences in proteomes influenced by lactation stages, parity, and exposure to tobacco smoke. The pilot study aimed to explore the feasibility of correlating certain proteins with several elements, considered as indicators of tobacco smoke and environmental influences on HBM.
