The peritrichous genus Ophrydium is relatively distinctive since its colonies have a gelatinous lorica. In the present work, one morphologically well-described species, Ophrydium crassicaule, was collected from a freshwater habitat in China. Both morphological characters and sequence data for SSU rDNA, ITS1-5.8S-ITS2 region and LSU rDNA were obtained. Morphologically, O. crassicaule is characterized by its elongated vase-shaped zooid, single-layered peristomial lip, dichotomously branched stalk, colony with gelatinous lorica, and infundibular polykinety 3 (P3) containing one short inner row and two long outer rows. Phylogenetic analyses revealed that the genus Ophrydium is monophyletic and nests within the family Vorticellidae. In addition, we briefly revise the nominal congeners for aiding the species identification in future studies.

Despite extensive previous morphological work, little agreement has been reached about phylogenetic relationships among peritrich ciliates, making it difficult to study the evolution of the group in a phylogenetic framework. In this study, the nucleotide characteristics and secondary structures of internal transcribed spacers 1 and 2 (ITS1 and ITS2) of 26 peritrich ciliates in 12 genera were analyzed. Information from secondary structures of ITS1 and ITS2 then was used to perform the first systematic study of ITS regions in peritrich ciliates, including one species of Rhabdostyla for which no sequence has been reported previously.

The phylogenetic relationships among peritrichs remain unresolved. In this study, the complete small subunit rRNA (SSrRNA) gene sequences of seven species (Epistylis galea, Campanella umbellaria, Carchesium polypinum, Zoothamnium arbuscula, Vaginicola crystallina, Ophrydium versatile, and Opercularia microdiscum) were determined. Trees were constructed using distance-matrix, maximum-likelihood and maximum-parsimony methods, all of which strongly supported the monophyly of the subclass Peritrichia.
