The histone methyltransferase SET domain bifurcated 1 (SETDB1) catalyzes the trimethylation of lysine 9 of histone H3, thereby regulating gene expression. In this study, we used conditional knockout mice, where Setdb1 was deleted only in neural crest cells (Setdb1 mice), to clarify the role of SETDB1 in palatal development. Setdb1 mice died shortly after birth due to a cleft palate with full penetration. Reduced palatal mesenchyme proliferation was seen in Setdb1 mice, which might be a possible mechanism of cleft palate development. Quantitative RT-PCR and in situ hybridization showed that expression of the Pax9, Bmp4, Bmpr1a, Wnt5a, and Fgf10 genes, known to be important for palatal development, were markedly decreased in the palatal mesenchyme of Setdb1 mice. Along with these phenomena, SMAD1/5/9 phosphorylation was decreased by the loss of Setdb1. Our results demonstrated that SETDB1 is indispensable for palatal development partially through its proliferative effect. Taken together with previous reports that PAX9 regulates BMP signaling during palatal development which implies that loss of Setdb1 may be involved in the cleft palate development by decreasing SMAD-dependent BMP signaling through Pax9.

Background: Orofacial cleft is among the most common craniofacial malformations. It presents a complex and multifactorial etiology that involves genetic and environmental factors. One of the etiological factors is consanguinity (marriage between blood relatives).

Background: Orofacial clefts are the most common craniofacial anomalies that include a variety of conditions affecting the lips and oral cavity. They remain a significant global public health challenge. Despite this, the quality of care for orofacial clefts has not been investigated at global and country levels.

Background: Early diagnosis of cleft lip and palate (CLP) requires a multiplane examination, demanding high technical proficiency from radiologists. Therefore, this study aims to develop and validate the first artificial intelligence (AI)-based model (CLP-Net) for fully automated multi-plane localization in three-dimensional(3D) ultrasound during the first trimester.
Methods: This retrospective study included 418 (394 normal, 24 CLP) 3D ultrasound from 288 pregnant woman between July 2022 to October 2024 from Shenzhen Guangming District People's Hospital during the 11-13 weeks of pregnancy.

Background/objectives: Evidence suggests nasal airflow resistance reduces after rapid maxillary expansion (RME). However, the medium-term effects of RME on upper airway (UA) airflow characteristics when normal craniofacial development is considered are still unclear. This retrospective cohort study used computer fluid dynamics (CFD) to evaluate the medium-term changes in the UA airflow (pressure and velocity) after RME in two distinct age-based cohorts.

Background: There are different intraoral appliances for cessation of thumb/finger sucking habit, but they have many disadvantages and to overcome it, extra oral appliances with colourful and attractive shape were developed. Electronic habit reminder in the form of wristwatch with alarming sound was assessed in cessation of thumb/finger sucking habit in children versus palatal crib after 6 and 9 months.
Methods: This study is a randomized clinical pilot study, with allocation ratio 1:1 parallel group.
