Background/objectives: Janus kinase inhibitors open new horizons for small-molecule drugs in treating inflammatory bowel disease, with ritlecitinib demonstrating significant efficacy in clinical trials for ulcerative colitis and Crohn's disease. Ritlecitinib, a second-generation JAK3 inhibitor, is a novel therapeutic agent for alopecia areata and other autoimmune conditions.

Methods: A new stability-indicating UHPLC-DAD-MS/MS method was developed, validated, and applied for a forced degradation study of ritlecitinib under ICH guidelines.

Psoriasis is a chronic inflammatory condition that is polygenic and multisystemic, impacting approximately 2-3% of the global population. : Janus kinase inhibitors (JAKi) have significantly advanced the treatment of various dermatological conditions by modulating the JAK-STAT signalling pathway. While these inhibitors have proven effective, they also pose challenges due to associated increase in serum lipid levels and relative potential cardiovascular risks. This perspective work aims to discuss the implications of these lipid alterations proposing management strategies for patients with dermatological disorders under JAKi treatments.

High-frequency ultrasound (HFUS) has been reported to be useful for the diagnosis of cutaneous diseases; however, its two-dimensional nature limits the value both in quantitative and qualitative evaluation. Three-dimensional (3D) visualization might help overcome the weakness of the currently existing HFUS. 3D-HFUS was newly developed and applied to various skin tumors and inflammatory hair diseases to assess its validity and advantages for dermatological use.

Alopecia areata (AA) is a non-scarring inflammatory hair loss condition associated with various immune-mediated comorbidities. Prediabetes, characterized by elevated blood glucose levels not yet high enough to be classified as diabetes, significantly increases the risk of developing type 2 diabetes mellitus (T2DM) and cardiovascular complications. The associations between AA obesity and prediabetes have long been investigated in an attempt to identify preventable risk factors, yet the literature is relatively scarce and inconclusive.
