Expanded carrier screening, a type of reproductive genetic testing for couples, has gained tremendous popularity for assessing the risk of passing on certain genetic conditions to offspring. Here, a carrier screening assay for 448 pathogenic variants was developed using capillary electrophoresis-based multiplex PCR technology. The capillary electrophoresis-based multiplex PCR assay achieved a sensitivity, specificity, and accuracy of 97.4%, 100%, and 99.6%, respectively, in detecting the specific variants. Among the 1915 couples (3830 individuals), 708 individuals (18.5%) were identified as carriers for at least one condition. Of the 708 carriers, 633 (89.4%) were heterozygous for one condition, 71 (10.0%) for two disorders, 3 (0.4%) for three disorders, and 1 (0.1%) for four disorders. Meanwhile, 30 (1.57%) couples were identified as at-risk couples. This study describes an inexpensive and effective method for expanded carrier screening. The simplicity and accuracy of this approach will facilitate the clinical implementation of expanded carrier screening.

COVID-19 outcomes are worse in non-pregnant patients that are cystic fibrosis carriers; however, no studies have examined COVID-19 outcomes in pregnant patients that are cystic fibrosis carriers. We evaluated the cystic fibrosis carrier status of pregnant patients with COVID-19 in three geographical regions in the United States and compared outcomes between non-carriers and carriers. Out of 2430 pregnant patients with COVID-19, 229 had a cystic fibrosis screen.

Recombinant human haemoglobin (rHb) is a tetramer protein with heme as cofactors, which have extensive applications in the fields of biomaterials and biomedical therapeutics. However, due to the poor structural stability, the dissociation of heme, weak oxygen transport efficiency, and lower activity, the utilisation of rHb is severely limited in artificial oxygen carriers. Herein, based on the novel developed high-throughput screening strategies and semi-rational design, the engineered rHb mutant with strong stability and heme-binding ability was obtained.

Li-Fraumeni syndrome (LFS) is an autosomal dominant cancer predisposition condition characterized by a high lifetime risk for a wide spectrum of malignancies associated with germline pathogenic/likely pathogenic (P/LP) variants in the TP53 tumor suppressor gene. Secondary malignant neoplasms are particularly common. Early cancer detection through surveillance enables early intervention and leads to improved clinical outcomes with reduced tumor-related mortality and treatment-related morbidity.
