Supramolecular hydrogels with a three-dimensional cross-linked macromolecular network have attracted growing scientific interest in recent years because of their ability to incorporate high loadings of bioactive molecules such as drugs, proteins, antibodies, peptides, and genes. Herein, we report a versatile approach for the confinement of lipase B (CALB) within a silica-strengthened cyclodextrin-derived supramolecular hydrogel and demonstrate its potential application in the selective oxidation of 2,5-diformylfuran (DFF) to 2,5-furandicarboxylic acid (FDCA) under mild conditions. The enzymatic nanoreactor was deeply characterized using thermogravimetric analysis, Fourier transform infrared spectroscopy, N-adsorption, dynamic light scattering, UV-visible spectroscopy, transmission electron microscopy, scanning electron microscopy, and confocal laser scanning microscopy, while the reaction products were established on the basis of H nuclear magnetic resonance spectroscopy combined with high-performance liquid chromatography. Our results revealed that while CALB immobilized in conventional sol-gel silica yielded exclusively 5-formylfuran-2-carboxylic acid (FFCA), confinement of the enzyme in the silicified hydrogel imparted a 5-fold increase in DFF conversion and afforded 67% FDCA yield in 7 h and almost quantitative yields in less than 24 h. The hierarchically interconnected pore structure of the host matrix was found to provide a readily accessible diffusion path for reactants and products, while its flexible hydrophilic-hydrophobic interface was extremely beneficial for the interfacial activation of the immobilized lipase.

This study aimed to investigate the impact of feeding patterns on the production performance, lipo-nutritional quality, and gut microbiota of Sunit sheep. A total of 24 sheep were assigned to two groups: confinement feeding (CF) and pasture feeding (PF) groups. After 90 days, the CF group exhibited significantly increased average daily gain, carcass weight, backfat thickness, and intramuscular fat content of the sheep, whereas the PF group showed significantly increased pH and decreased L value and cooking loss of the longissimus lumborum (LL) muscle (P < 0.

Enzymes are known for their remarkable catalytic efficiency across a wide range of applications. Here, we present a novel and convenient nanoreactor platform based on zwitterionic polyelectrolyte complex vesicles (PCVs), assembled from oppositely charged homopoly(2-oxazoline)s, facilitating enzyme immobilization. We show remarkable enhancements in catalytic activity and stability by encapsulation of lipase as a model enzyme.

Harnessing the power of cell biocatalysis for sustainable chemical synthesis requires rational integration of living cells with the modern synthetic catalysts. Here, we develop silica-tiling strategy that constructs a hierarchical, inorganic, protocellular confined nanospace around the individual living cell to accommodate molecularly accessible abiotic catalytic sites. This empowers the living microorganisms for new-to-nature chemical synthesis without compromising the cellular regenerative process.

Spatial immobilization of fragile enzymes using a nanocarrier is an efficient means to design heterogeneous biocatalysts, presenting superior stability and recyclability to pristine enzymes. An immobilized enzyme, however, usually compromises its catalytic activity because of inevasible mass transfer issues and the unfavorable conformation changes in a confined environment. Here, we describe a synergetic metal-organic framework pore-engineering strategy to trap lipase (an important hydrolase), which confers lipase-boosted stability and activity simultaneously.
