Amphiphilic polyurethane elastomers (APUE) were synthesized using a two-step polyaddition reaction based on the hydroxyl-terminated polydimethylsiloxane (PDMS) and polyethylene glycol (PEG) soft segments with the molecular weights ('s) of 2000 and 1000, respectively. The effects of the PDMS/PEG contents on the properties and structures of the APUE were investigated. It was found that the APUE possessed high elongation, moderate tensile strength, and good thermal properties. In addition, the APUE showed tunable oxygen permeability (Dk) and water vapor transmission rate (WVTR), and a similar WVTR to that of skin could be obtained for the optimized sample (APUE2). Importantly, APUE also exhibited excellent antibacterial efficacy against two kinds of bacteria along with impressive cytocompatibility. All of the results demonstrated that the synthesized APUE will hold substantial potential for biomaterial applications.

Modulating microbial motility and physiology can enhance the production of bacterial macromolecules and small molecules. Herein, a platform of water-soluble and amphiphilic peptidomimetic polyurethanes is reported as a means of regulating bacterial surface behavior and the concomitant production of extracellular polymeric substances (EPS). It is demonstrated that carboxyl (-COOH)-containing polyurethanes exhibited 17-fold and 80-fold enhancements in () swarming and twitching areas, respectively.

This paper reports synthesis, aqueous self-assembly and relevance of the pH-triggered activable photodynamic therapy of amphiphilic polyurethane (P1S) functionalized with a heavy-atom free organic photosensitizer. Condensation polymerization between 1,4-diisocyanatobutane and two different dihydroxy monomers (one having a pendant hydrophilic wedge and the other having 1,3-dihydroxypropan-2-one with a reactive carbonyl group) in the presence of a measured amount of ()-2-methylbutan-1-ol (chain-stopper) and DABCO catalyst produces a reactive pre-polymer P1. Hydrazide-functionalized thionated-naphthalenemonoimide (NMIS), which acts as a photosensitizer, reacted with the carbonyl-functionality of P1 to obtain the desired polymer-photosensitizer conjugate P1S in which the dye was attached to the polymer backbone an acid-labile hydrazone linker.

To synthesize an effective and versatile nano-platform serving as a promising carrier for controlled drug delivery, visible-light-induced diselenide-crosslinked polyurethane micelles were designed and prepared for ROS-triggered on-demand doxorubicin (DOX) release. A rationally designed amphiphilic block copolymer, poly(ethylene glycol)--poly(diselenolane diol-co-isophorone diisocyanate)--poly(ethylene glycol) (PEG--PUSe--PEG), which incorporates dangling diselenolane groups within the hydrophobic PU segments, was initially synthesized through the polycondensation reaction. In aqueous media, this type of amphiphilic block copolymer can self-assemble into micellar aggregates and encapsulate DOX within the micellar core, forming DOX-loaded micelles that are subsequently in situ core-crosslinked by diselenides via a visible-light-triggered metathesis reaction of Se-Se bonds.
