Terahertz (THz) radiation has attracted considerable attention in medical imaging owing to its nonionizing and spectral fingerprinting characteristics. To date, most studies have focused on in vitro and ex vivo objects with water-removing pretreatment because the water in vivo excessively absorbs the THz waves, which causes deterioration of the image quality. In this review, we discuss how THz medical imaging can be used for a living body. The development of imaging contrast agents has been particularly useful to this end. In addition, we also introduce progress in novel THz imaging methods that could be more suitable for in vivo applications. Based on our discussions, we chart a developmental roadmap to take THz medical imaging from in vitro to in vivo.

Background: Normal brain aging is associated with dopamine decline, which has been linked to age-related cognitive decline. Factors underlying individual differences in dopamine integrity at older ages remain, however, unclear. Here we aimed at investigating: (i) whether inflammation is associated with levels and 5-year changes of in vivo dopamine D2-receptor (DRD2) availability, (ii) if DRD2-inflammation associations differ between men and women, and (iii) whether inflammation and cerebral small-vessel disease (white-matter lesions) serve as two independent predictors of DRD2 availability.

The most well-studied approach to enhance brain influx of protein therapeutics, is receptor-mediated transcytosis (RMT) by targeting nutrient receptors to shuttle protein therapeutics over the blood-brain barrier (BBB) along with their endogenous cargos. While higher brain exposure is achieved with RMT, the timeframe is short due to rather fast brain clearance.

Background: Optical spectral transmission (OST) is a modern diagnostic method capable of quantifying inflammation in the finger and wrist joints of arthritis patients by assessing the blood-specific absorption of light transmitted through a tissue.
