This study aimed to investigate the highly differentiated urothelial apical surface glycome. The functions of the mammalian urothelium, lining the majority of the urinary tract and providing a barrier against toxins in urine, are dependent on the correct differentiation of urothelial cells, relying on protein expression, modification, and complex assembly to regulate the formation of multiple differentiated cell layers. Protein glycosylation, a poorly studied aspect of urothelial differentiation, contributes to the apical glycome and is implicated in the development of urothelial diseases. To enable surface glycome characterization, we developed a method to collect tissue apical surface - and O-glycans. A simple, novel device using basic laboratory supplies was developed for enzymatic shaving of the luminal bladder urothelial surface, with subsequent release and mass spectrometric analysis of apical surface O- and -glycans, the first normal mammalian urothelial -glycome to be defined. Trypsinization of superficial glycoproteins was tracked using immunolabeling of the apically expressed uroplakin 3a protein to optimize enzymatic release, without compromising the integrity of the superficial urothelial layer. The approach developed for releasing apical tissue surface glycans allowed for comparison with the -glycome of the total porcine bladder urothelial cells and thus identification of apical surface glycans as candidates implicated in the urothelial barrier function. One area of technological advancement has been the shift from stainless steel hand tools to nickel-titanium (Ni-Ti) rotary tools. This paper aims to perform an in vitro comparative study to evaluate the efficacy of five endodontic manual and rotary instruments such as Kerr files, Orodeka Plex V, ProTaper Flydent NiTi super files, and ProTaper Flydent NiTi super files in combination with an ultrasonic endodontic E3D Diamantata EMS scaler used for root canal shaping. The following aspects were highlighted: effective removal of smear layer (SL) from the dentinal tubules in the coronal 1/3, middle 1/3, and apical 1/3 of the root canal, appearance of cracks in the dentinal walls by SEM analysis, and highlighting of dentin mineral content and remnant debris by EDX analysis.

Aim To compare the quality of obturation using WVC (warm vertical compaction), CLC (cold lateral compaction), injectable (iFill), and Thermafil (GuttaCore) techniques, along with hydraulic condensation of bioceramic (BC) sealer, and evaluating the percentage of gutta-percha (GP), sealer, and voids in simulated internal resorptive lesions, using an advanced stereomicroscope with ImageJ software (National Institutes of Health, Bethesda, MD, USA). Methods and material In this study, 40 freshly extracted maxillary incisors were collected, and endodontic instrumentation was done to working length using hand K-files. Simulated internal resorption cavities were created in the middle-apical third of the roots after horizontal sectioning and were re-cemented.
