TRPM2 is a calcium permeable non-selective cation channel involved in many important physiological processes and has divergent gating mechanisms across species. Structural studies have revealed that TRPM2 is gated by adenosine 5'-diphosphoribose that binds to the cytosolic domains of TRPM2 and calcium ions that are coordinated by residues in the transmembrane domain. However, the selectivity filter of human TRPM2 remains elusive due to the poor resolution in this region. In a recent manuscript published in Cell Reports, Yu et al. present unexpected dual roles of the selectivity filter in human TRPM2 by determining a high-resolution structure of human TRPM2 in lipid nanodiscs. This study provides unprecedented insights into the gating mechanism of human TRPM2.

Background: The factors contributing to the development of severe coronavirus disease 2019 (COVID-19) following infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) remain unclear. Although the presence of immune complexes (ICs), formed between antibodies and their antigens, has been linked to COVID-19 severity, their role requires further investigation, and the antigens within these ICs are yet to be characterized.

Method: Here, a C1q enzyme-liked immunosorbent assay and immune complexome analysis were used to determine IC concentrations and characterize IC antigens, respectively, in the sera of 64 unvaccinated COVID-19 patients with PCR-confirmed SARS-CoV-2 infection, enrolled at seven participating centers in 2020.

Psoriatic arthritis (PsA) and rheumatoid arthritis (RA) are connective tissue autoimmune diseases. The present study aimed to check whether serum clusterin (CLU) concentration and its glycosylation pattern may be markers differentiating these diseases-blood sera of patients with PsA (n = 37), RA (n = 34), and healthy subjects (control, n = 21) were examined.
