This research article reports the electrical detection of breast-cancer biomarker (C-erbB-2) in saliva/serum based on In1-xGaxAs/Si heterojunction dopingless tunnel FET (HJ-DL-TFET) biosensor for highly sensitive and real-time detection. The work takes into account the interface charge modulation effect in dopingless extended gate heterostructure TFET with embedded nanocavity biosensors for the precise, reliable, and fast detection of antigens present in the body fluids such as saliva in place of blood serum. The reported biosensor is numerically simulated in 2D using the SILVACO ATLAS exhaustive calibrated simulation framework. For the biomolecule immobilization, the proposed biosensor has a dual cavity engraved beneath the dual gate structure. This improves the control of biomolecules over the source-to-channel tunneling rate, as well as the control over the electrical performance parameters of the proposed biosensor. Here, a numerical model for the C-erbB-2 interface charge equivalent is also developed. The analysis of device sensitivity in both saliva and serum environments for various C-erbB-2 concentrations has been carried out. Our study reveals that III-V In1-xGaxAs/Si heterojunction with x composition of 0.2 and extended gate geometry provides an increased tunneling probability, improves the gate control to get a higher ION/IOFF ratio and higher sensitivity. In addition to this, the impact of interface charges corresponding to the different amounts of C-erbB-2 biomarkers on the biosensor sensitivity (in terms of ION/IOFF ratio) yields higher sensitivity of the order of 106.

Introduction: Breast cancer (BC) is the most prevalent malignant tumor in women, with triple-negative breast cancer (TNBC) showing the poorest prognosis among all subtypes. Glycosylation is increasingly recognized as a critical biomarker in the tumor microenvironment, particularly in BC. However, the glycosylation-related genes associated with TNBC have not yet been defined.

MicroRNAs (miRNAs) are pivotal biomarkers for cancer screening. Identifying distinctive expression patterns of miRNAs in specific cancer types can serve as an effective strategy for classification and characterization. However, the development of a minimal signature of miRNAs for accurate cancer classification remains challenging, hindered by the lack of integrated approaches that systematically analyse miRNA expression levels of miRNAs alongside their associated biological pathways.

Triple-negative breast cancer (TNBC) is a unique breast cancer subtype characterized by the lack of estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor 2 (HER2) expression in tumor cells. TNBC represents about 15% to 20% of all breast cancers and is aggressive and highly malignant. Currently, TNBC diagnosis primarily depends on pathological examination, while treatment efficacy is assessed through imaging, biomarker detection, pathological evaluation, and clinical symptom improvement.

ZNF165: A Pan-Cancer Biomarker with Prognostic and Therapeutic Potential.

Protein Pept Lett

January 2025

Scientific Research Center, Beijing ChosenMed Clinical Laboratory Co., Ltd. 101, 1F, Building 3, No.156 Jinghai 4th Road, Beijing Economic and Technological Development Zone, Beijing, 100176, China.

Background: The role of ZNF165 in only a few tumors has been reported. ZNF165 plays an important role in liver cancer, gastric cancer, and breast cancer, especially in regulating the immune microenvironment, promoting tumor cell proliferation and migration, and serving as a potential target for immunotherapy.

Objective: This study aimed to enhance an understanding of how the ZNF165 gene functions and influences cancer development.

Triple-negative breast cancer (TNBC) remains a significant global health challenge, emphasizing the need for precise identification of patients with specific therapeutic targets and those at high risk of metastasis. This study aimed to identify novel therapeutic targets for personalized treatment of TNBC patients by elucidating their roles in cell cycle regulation. Using weighted gene co-expression network analysis (WGCNA), we identified 83 hub genes by integrating gene expression profiles with clinical pathological grades.
