To investigate the role of sulfhydryl groups and disulfide bonds in different protein-stabilized emulsions, -ethylmaleimide (NEM) was used as a sulfhydryl-blocking agent added in the emulsion. The addition of NEM to block the sulfhydryl groups resulted in a reduction in disulfide bond formation, which enabled the internal structure of the protein molecule to be destroyed, and then decreased the restriction of protein membrane on the oil droplets. Furthermore, with the NEM content increasing in the emulsion, a reduction in the protein emulsifying activity and emulsion stability also occurred. At the same time, the intermolecular interaction of the protein on the oil droplet interface membrane was destroyed, and the emulsion droplet size increased with the NEM content in the emulsion. Although NEM blocking sulfhydryl groups from forming disulfide bonds has similar effects on three types of protein emulsion, the degree of myofibrillar protein (MP), egg-white protein isolate (EPI), and soybean protein isolate (SPI) used as emulsifiers had a subtle difference.

We previously used high pressure liquid chromatography (HPLC) coupled with Se-specific inductively coupled plasma mass spectrometry (ICP-MS) and molecule specific (ESI Orbitrap MS/MS) detection to study the increase in liver Se in turkeys and rats supplemented as selenite in high-Se (5 µg Se/g diet) and adequate-Se diets. We found that far more Se is present as selenosugar (seleno-N-acetyl galactosamine) than is present as selenocysteine (Sec) in true selenoproteins. In high-Se liver, the increase in liver Se was due to low molecular weight (LMW) selenometabolites as glutathione-, cysteine- and methyl-conjugates of the selenosugar, but also as high molecular weight (HMW) species as selenosugars decorating general proteins via mixed-disulfide bonds.

Intracellular delivery of proteins is an important barrier in the development of strategies to deliver functional proteins and protein therapeutics into the cells to realize their full potential in biotechnology, biomedicine, cell-based therapies, and gene editing protein systems. Cysteine derivatives having disulfide bonds in their side chains can be used as redox-responsive organogelators. The disulfide bond can be cleaved in the presence of certain reducing agents like thiol derivatives such as glutathione (GSH), which is a tripeptide that consists of cysteine, glutamic acid, and glycine. To understand the effects of compound curing agent (60 % NaCl+40 % KCl + 90 mg/kg NaNO) on the protein characteristics and proteome of Nuodeng ham, the protein structure, oxidation, degradation, and proteomic changes of Nuodeng ham were analyzed. The results showed that the hydrophobic and disulfide bonds were the main forces to maintain the stability of the ham protein. The compound curing agent reduced protein oxidation and promoted the transformation of the α-helix structure to the β-sheet structure.
