Traumatic brain injury (TBI) precipitates a neuroinflammatory cascade, with the NLRP3 inflammasome emerging as a critical mediator. This review scrutinizes the complex activation pathways of the NLRP3 inflammasome by underscoring the intricate interplay between calcium signaling, mitochondrial disturbances, redox imbalances, lysosomal integrity, and autophagy. It is hypothesized that a combination therapy approach-integrating NF-κB pathway inhibitors with NLRP3 inflammasome antagonists-holds the potential to synergistically dampen the inflammatory storm associated with TBI.

In vitro and in vivo effects of mesoporous silica nanoparticles (MSN) on the functional activity of platelets were studied in experiments on white rats. MSN particles, neither uncoated nor coated with calcium alginate, induced spontaneous platelet aggregation when added to platelet-rich plasma, but significantly enhanced ADP-induced platelet aggregation. Subcutaneous administration of uncoated and calcium alginate-coated MSN resulted in increased maximum size and rate of platelet aggregate formation 1 day post-injection.

Cocaine abstinence and withdrawal are linked to relapse, heightened anxiety, and depressive-like symptoms. While L-type calcium channels (LTCCs) have been associated with cocaine use disorders in humans and drug-seeking behavior in rodent models, their role in mood-related symptoms during cocaine abstinence remains unclear. This study examined whether blocking LTCCs with isradipine could alter anxiety and depressive symptoms induced by cocaine abstinence.
