Inhibition of the RNA-binding protein LIN28 and disruption of the protein-RNA interaction of LIN28-let-7 with small molecules holds great potential to develop new anticancer therapeutics. Herein, we report the LIN28 inhibitory activities of a series of 30 small molecules with a tricyclic tetrahydroquinoline (THQ)-containing scaffold obtained from a Povarov reaction. The THQ molecules were structurally optimized by varying the 2-benzoic acid substituent, the fused ring at 3- and 4-positions, and the substituents at the phenyl moiety of the tetrahydroquinoline core. Among the tested compounds, GG-43 showed dose-dependent inhibition in an EMSA validation assay and low micromolar inhibitory activity in a fluorescence polarization-based assay measuring disruption of LIN28-let-7 interaction. Binding mode between GG-43 and the cold shock domain of LIN28 was proposed via a molecular docking analysis. The study provides one of the first systematic analyses on structural features that are required for LIN28 inhibition, and indicates the necessity to develop small molecules with new scaffolds as LIN28-targeting probes and therapeutic candidates. In parallel, this study demonstrates the polypharmacological nature of tricyclic THQ-containing scaffolds accessible through Povarov reactions.

Gold-catalyzed synthesis of decorated indoles has been developed through carbene insertion into N-H bonds of -allyl-2-(aryl/alkyl ethynyl)anilines using α-diazo compounds followed by cyclization and concomitant 1,3 migration of allyl fragments. The developed protocol tackles the inherited challenge of direct C3 functionalization and eliminates the need for a tertiary aniline precursor for the 1,3-migration reaction. The applicability of this transformation is showcased through the practical synthesis of analogs of small drug-like and pharmaceutically relevant molecules such as ibuprofen, estradiol, menthol, and borneol, The mechanism is well supported by control experiments and isolation of the reaction intermediate.

In this study, the first attempt is made to implement conjugated polymer-based self-assembled monolayer (SAM), poly[3-(6-carboxyhexyl) thiophene-2,5-diyl] (P3HT-COOH), is implemented as the hole transport layer (HTL) in fabricatiing organic photovoltaics (OPVs). The scanning tunneling microscopy (STM) results show that those P3HT-COOH molecules with periodic carboxylic acid anchoring groups pack periodically on the indium tin oxide (ITO) surface and form a monolayer. Further, this monolayer is smooth and dense with a polar feature that minimizes defects, forms an excellent interface with the photoactive layer, and tunes the work function of ITO beneficial for hole extraction.

Overcoming the challenges of integrating disparate components in nanoarchitectures, this study introduces a straightforward strategy based on a mixed-valence coordination approach, creating an ordered ternary heterostructure integrated with ultrasmall homojunction. This singular ordered homojunction-heterostructure unites ultrathin 1D rutile TiO nanowires (NWs) and ultrathin anatase TiO NWs with 0D Prussian Blue Analogs (PBAs) nanoparticles (NPs), all exhibiting crystallographic oriented alignment with each other, forming a ternary mesocrystals. Experimental and theoretical insights disclose that the complex interplay between these dissimilar components is governed by a spontaneous lattice match effect, which not only optimizes but also directs the charge transfer, thereby enhancing both efficiency and stability.

Background: The specific and non-specific toxicities of cryoprotective agents (CPAs) for semen or spermatozoa cryopreservation/vitrification (SC/SV) remain challenges to the success of assisted reproductive technologies.
Objective: We searched for and integrated the physicochemical and toxicological characteristics of small-molecule CPAs as well as curated the information of all extenders reported for carnivores to provide a foundation for new research avenues and computational cryobiology.
Methods: The PubMed database was systematically searched for CPAs reported in SC/SV of carnivores from 1964 to 2024.
