Background: Laboratory diagnosis of Lyme borreliosis refers to some methods with known limitations. Molecular diagnostics using specific nucleic acid probes may overcome some of these limitations.
Methods: We describe the novel reporter fluorescence real-time polymerase chain reaction (PCR) probe system for detection of species. Advantages of the system include having cheap conventional fluorescence dyes, easy primer design, no restrictions for PCR product lengths, robustness, high sequence specificity, applicability for multiplex real-time PCRs, melting curve analysis (single nucleotide polymorphism analysis) over a large temperature range, high sensitivity, and easy adaptation of conventional PCRs.
Results: Using the probe system we were able to detect all nine tested European species belonging to the (sensu lato) complex and differentiated them from relapsing fever species. As few as 10 copies of in one PCR reaction were detectable.
Conclusion: We established a novel multiplex probe real-time PCR system, designated , that is simple, robust, and incorporates melting curve analysis for the detection and in the differentiation of European species belonging to the s.l. complex.

Knowledge of protein-metabolite interactions can enhance mechanistic understanding and chemical probing of biochemical processes, but the discovery of endogenous ligands remains challenging. Here, we combined rapid affinity purification with precision mass spectrometry and high-resolution molecular docking to precisely map the physical associations of 296 chemically diverse small-molecule metabolite ligands with 69 distinct essential enzymes and 45 transcription factors in the gram-negative bacterium Escherichia coli. We then conducted systematic metabolic pathway integration, pan-microbial evolutionary projections, and independent in-depth biophysical characterization experiments to define the functional significance of ligand interfaces.

As a zoonotic virus, highly sensitive detection of monkeypox virus is crucial for its prevention and control due to its rapid increase in cases worldwide and the extremely high risk of virus transmission. In this paper, based on the principle of antigen-antibody specific recognition, an ultrasensitive resonance Raman biosensing probe was prepared using a molecule with the bifunctionality of resonance Raman effect and capturing antibody; and with the strong affinity of the biotin-streptavidin (Bio-SA) system, Bio-antibody and SA test strips were prepared.
