Several phenanthrenes (), phenolics () and steroidal sapogenins () were isolated for the first time from the aqueous and methanolic extracts of Pax yam collected from Tanzania. Chemical structures of all the isolates () were determined by using 1D and 2D nuclear magnetic resonance spectral methods. All pure isolates were evaluated for anti-inflammatory activity using cyclooxygenase enzyme (COX-1 and -2) inhibitory assays. Among the isolates tested, phenanthrenes showed the highest COX-1 and -2 enzyme inhibitory activity whereas phenolics () and steroidal sapogenins () exhibited moderate inhibition when compared to non-steroidal anti-inflammatory drugs aspirin, ibuprofen and naproxen. Compounds were evaluated for antioxidant activity using lipid peroxidation inhibitory (LPO) assay for the first time and exhibited moderate LPO inhibition.

Activating the pyroptosis pathway of tumor cells by photodynamic therapy (PDT) for immunogenic cell death (ICD) is considered a valid strategy in pursuit of antitumor immunotherapy, but it remains a huge challenge due to the lack of reliable design guidelines. Moreover, it is often overlooked that conventional PDT can exacerbate the development of tumor immunosuppressive microenvironment, which is apparently unfavorable to clinical immunotherapy. The endoplasmic reticulum's (ER) pivotal role in cellular homeostasis and its emerging link to pyroptosis have galvanized interest in ER-centric imaging and therapeutics.

Background: The thromboxane A2 receptor (TPαR) plays an important role in the amplification of platelet responses during thrombosis. Receptor activity is regulated by internalization and receptor desensitization. The mechanism by which constitutive surface expression of the TPαR is regulated is unknown.

Cyclooxygenase-2 (COX-2) is an enzyme that plays a crucial role in inflammation by converting arachidonic acid into prostaglandins. The overexpression of enzyme is associated with conditions such as cancer, arthritis, and Alzheimer's disease (AD), where it contributes to neuroinflammation. In silico virtual screening is pivotal in early-stage drug discovery; however, the absence of coding or machine learning expertise can impede the development of reliable computational models capable of accurately predicting inhibitor compounds based on their chemical structure.

Inflammation is a physiological response of the immune system to infectious agents or tissue injury, which involves a cascade of vascular and cellular events and the activation of biochemical pathways depending on the type of harmful agent and the stimulus generated. The Kunitz peptide HCIQ2c1 of sea anemone is a strong protease inhibitor and exhibits neuroprotective and analgesic activities. In this study, we investigated the anti-inflammatory potential of HCIQ2c1 in histamine- and lipopolysaccharide (LPS)-activated RAW 264.
