Circulating MicroRNAs in Idiopathic Pulmonary Fibrosis: A Narrative Review.

Curr Issues Mol Biol

December 2024

Molecular Biology Laboratory, Department of Research in Pulmonary Fibrosis, National Institute of Respiratory Diseases "Ismael Cosío Villegas", Calzada de Tlalpan 4502, Col. Sección XVI, Mexico City 14080, Mexico.

Idiopathic pulmonary fibrosis (IPF) is a chronic, deathly disease with no recognized effective cure as yet. Furthermore, its diagnosis and differentiation from other diffuse interstitial diseases remain a challenge. Circulating miRNAs have been measured in IPF and have proven to be an adequate option as biomarkers for this disease.

Article Synopsis
  • - The study highlights the connection between alcohol consumption and lung cancer, focusing on the inactivating mutation in the ALDH2 enzyme prevalent in Asian populations, which impacts acetaldehyde clearance.
  • - Through a bioinformatic analysis of multi-omics data, researchers found that ALDH2 is significantly expressed in healthy lung tissue but less so in lung adenocarcinoma (LUAD), affecting key cellular processes and correlating with poorer patient prognosis.
  • - The findings suggest that understanding ALDH2's role in lung cancer could lead to its use as a prognostic marker and a target for therapies aimed at enhancing immune responses in LUAD patients.

Background: Bronchial asthma, the most prevalent chronic inflammatory airway disease in children, exhibits a concerning rise in both incidence and prevalence. Asthma biomarkers hold promise for stratifying patients into distinct clinical phenotypes, paving the way for targeted and personalized treatment approaches.

Aim Of Study: This study aimed to evaluate the association between novel and non-established semi-invasive circulating and well-known exhaled inflammatory biomarkers in two distinct pediatric asthma populations stratified by disease severity.

Inhaled therapy has become a crucial treatment option for respiratory diseases like asthma, cystic fibrosis, and chronic obstructive pulmonary disease (COPD), delivering drugs directly to bronchial and alveolar tissues. However, traditional static cell models, while valuable for studying pharmacokinetics (PK) and pharmacodynamics (PD), fall short in replicating the dynamic nature of physiological breathing. In this study, we present a breathing lung chip model that integrates a dynamic breathing mechanism with an air-liquid interface (ALI) culture environment to overcome these limitations.

Purpose: Poor lung cancer patients' outcomes and survival rates demand the discovery of new biomarkers for the specific, significant, and less invasive detection of non-small cell lung cancer (NSCLC) progression. The present study aimed to investigate the potential of miRNA expression as biomarkers in NSCLC utilizing a preclinical cell culture setup based on screening of miRNAs in NSCLC cells grown in 3D cell culture.

Patients And Methods: The study was performed using lung cancer cell lines, varying in different levels of aggressiveness: NCI-H1299, A549, Calu-1, and NCI-H23, as well as noncancerous bronchial epithelial cell line HBEC3, which were grown in 3D cell culture.
