Major progress has been made in defining the basis of the mitochondrial permeability transition, a Ca -dependent permeability increase of the inner membrane that has puzzled mitochondrial research for almost 70 years. Initially considered an artefact of limited biological interest by most, over the years the permeability transition has raised to the status of regulator of mitochondrial ion homeostasis and of druggable effector mechanism of cell death. The permeability transition is mediated by opening of channel(s) modulated by matrix cyclophilin D, the permeability transition pore(s) (PTP). The field has received new impulse (a) from the hypothesis that the PTP may originate from a Ca -dependent conformational change of F-ATP synthase and (b) from the reevaluation of the long-standing hypothesis that it originates from the adenine nucleotide translocator (ANT). Here, we provide a synthetic account of the structure of ANT and F-ATP synthase to discuss potential and controversial mechanisms through which they may form high-conductance channels; and review some intriguing findings from the wealth of early studies of PTP modulation that still await an explanation. We hope that this review will stimulate new experiments addressing the many outstanding problems, and thus contribute to the eventual solution of the puzzle of the permeability transition.

Tumor Microenvironment-Driven Structural Transformation of Vanadium-Based MXenzymes to Amplify Oxidative Stress for Multimodal Tumor Therapy.

Adv Sci (Weinh)

National Engineering Research Center for Biomaterials, College of Biomedical Engineering, Sichuan University, Chengdu, 610064, China.

MXenzymes, a promising class of catalytic therapeutic material, offer great potential for tumor treatment, but they encounter significant obstacles due to suboptimal catalytic efficiency and kinetics in the tumor microenvironment (TME). Herein, this study draws inspiration from the electronic structure of transition metal vanadium, proposing the leverage of TME specific-features to induce structural transformations in sheet-like vanadium carbide MXenzymes (TVMz). These transformations trigger cascading catalytic reactions that amplify oxidative stress, thereby significantly enhancing multimodal tumor therapy.

Mitochondrial homeostasis is crucial for maintaining cellular energy production and preventing oxidative stress, which is essential for overall cellular function and longevity. Mitochondrial damage and dysfunction often occur concomitantly in myocardial ischemia-reperfusion injury (MIRI). Notoginsenoside R1 (NGR1), a unique saponin from the traditional Chinese medicine Panax notoginseng, has been shown to alleviate MIRI in previous studies, though its precise mechanism remains unclear.

Composite adsorbents based on a natural biopolymer matrix of chitosan, to which 4-amino-N'-hydroxy-1,2,5-oxadiazole-3-carboximidamide and its Se derivative were attached, were synthesized. A complex of physicochemical analysis methods indicates that the direct introduction of a matrix with high ionic permeability into the reaction mixture contributes to the formation of homogeneous particles of composite with developed surface morphology, which enhances the kinetic and capacitive parameters of uranium sorption in liquid media. It has been established that the direct introduction of a matrix with high ionic permeability into the reaction mixture contributes to the formation of homogeneous particles with a developed surface morphology, which enhances the kinetic and capacitive parameters of uranium sorption in liquid media.

The gas (or plastron) trapped between micro/nano-scale surface textures, such as that on superhydrophobic surfaces, is crucial for many engineering applications, including drag reduction, heat and mass transfer enhancement, anti-biofouling, anti-icing, and self-cleaning. However, the longevity of the plastron is significantly affected by gas diffusion, a process where gas molecules slowly diffuse into the ambient liquid. In this work, we demonstrated that plastron longevity could be extended using a gas-soluble and gas-permeable polydimethylsiloxane (PDMS) surface.
