AI Article Synopsis

  • Three perylene bisimide-glycopeptide conjugates were created, showing moderate effectiveness in controlling ice crystal growth.
  • These conjugates, particularly PBI-AFF-Glu, significantly enhanced cell viability during cryopreservation, achieving 22.77% for HeLa cells, 19.43% for A549 cells, and 16.63% for GES-1 cells at a concentration of 1.0 mg/mL.
  • The findings aim to aid in the future development of self-assembled cryoprotectants for preserving cells.

Article Abstract

Three perylene bisimide-glycopeptide conjugates (PBI-AFF-Man, PBI-AFF-Glu and PBI-AFF-Gal) were synthesized, which showed moderate activity in the control of ice crystal growth. Furthermore, the cellular cryopreservation effects of PBI-AFF-Man, PBI-AFF-Glu and PBI-AFF-Gal showed enhancements in cell viabilities, especially for PBI-AFF-Glu with values of 22.77 ± 3.33% (HeLa cells), 19.43 ± 1.90% (A549 cells) and 16.63 ± 1.76% (GES-1 cells) at a dose of 1.0 mg mL. This work will help guide the development of self-assembled cryoprotectants.

Gut microbiota disruptions after allogeneic hematopoietic cell transplantation (alloHCT) are associated with increased risk of acute graft-versus-host disease (aGVHD). We designed a randomized, double-blind placebo-controlled trial to test whether healthy-donor fecal microbiota transplantation (FMT) early after alloHCT reduces the incidence of severe aGVHD. Here, we report the results from the single-arm run-in phase which identified the best of 3 stool donors for the randomized phase.

The application of spermatogonial stem cells (SSC) will be more effective and feasible following the successful cryopreservation and transfer of SSCs in livestock. Like other cells, SSCs are also sensitive to cryoinjury; hence composition of the cryomedia and freezing protocols need to be optimized. The present study aims to optimising the best freezing rates by minimising the ice crystallization and dehydration effect in order to maximize the post-thaw SSCs survivability and stemness characteristics.

Platelet-Rich Plasma (PRP) is a biological treatment widely used in regenerative medicine for its restorative capacity. Although PRP is typically applied at the time of obtention, long-term storage and preservation could enhance its versatility and clinical applications. The objective of this study was to evaluate the effect of long-term freezing on PRP.

Cryopreservation can adversely affect sperm motility, structural integrity, and fertilization ability. This study investigated the effects of MitoQ and antifreeze protein III (AFP III) on frozen-thawed semen from eight adult dogs using a Tris-fructose extender. Ejaculates were divided and diluted with a standard Tris-fructose-egg yolk extender containing MitoQ (200 nM/mL) and AFP III (0.

Due to oxidative damage and mitochondrial dysfunction, boar semen cryopreservation remains a significant challenge. This study investigates the effects of pyrroloquinoline quinone (PQQ), a mitochondrial-targeted antioxidant, on the post-thaw boar sperm quality during cryopreservation. Boar semen was diluted in a freezing extender containing different concentrations of PQQ (0, 10, 100, 1000, 10,000 nM).
