Fourteen polyhydroxylated steroids including six new compounds, ehrensteroids A - F (1-6), were isolated from the Vietnamese soft coral Sarcophyton ehrenbergi after applying various chromatographic separations. The structure elucidation was done by detailed analysis of 1D, 2D NMR and HR QTOF mass spectra. In addition, significant cytotoxicity (IC values ranging from 8.45 ± 0.56 to 38.27 ± 2.39 μM) against all five tested cancer cell lines as LU-1 (lung cancer), HepG2 (hepatoma cancer), MCF-7 (breast cancer), HL-60 (acute leukemia), and SW480 (colon adenocarcinoma) was observed for compounds 1, 2, 6-10, and 12-14, whereas this activity on LU-1 (IC = 21.91 ± 2.72 μM) and HepG2 (IC = 20.32 ± 0.03 μM) cell lines was also observed for 4.

20-hydroxyecdysone (20E), a natural polyhydroxylated steroid, has exhibited anti-inflammatory effects across various diseases. This study investigates the potential connection between 20E's anti-inflammatory properties and the RNA-binding protein Lin28b, which is notably upregulated in TNF-α-stimulated endothelial cells. Herein, we discovered that 20E can selectively downregulate Lin28b expression without affecting its paralog Lin28a.

Insect Sterols and Steroids.

Adv Exp Med Biol

BIOSIPE, Sorbonne Université, Paris, France.

Insects are incapable of biosynthesising sterols de novo so they need to obtain them from their diets or, in certain cases, from symbiotic microorganisms. Sterols serve a structural role in cellular membranes and act as precursors for signalling molecules and defence compounds. Many phytophagous insects dealkylate phytosterols to yield primarily cholesterol, which is also the main sterol that carnivorous and omnivorous insects obtain in their diets.

Polyhydroxylated spirostanol saponins, characterized by three or more hydroxy substitutions in the aglycone, have various interesting biological activities. In the present study, "steroids", "saponins", "polyhydroxylated", "spirostanol saponins", and "steroidal saponins" were used as search terms to screen the literature. Cited references were collected between 1950 and 2023 from the Web of Science, SciFinder, and China National Knowledge Internet (CNKI).

Article Synopsis
  • Ecdysteroids are a class of anabolic steroids that function as important hormones in insects, but their analysis is challenging due to low levels in biological samples and fluctuations during development.
  • A new method has been designed to accurately quantify four specific ecdysteroids after extraction and conversion to enhance detectability, enabling measurement from minimal biological material.
  • The method has been validated using firebug larval hemolymph and successfully applied to other biological samples, including human serum, showcasing its versatility and effectiveness in detecting ecdysteroids.
