Stem cells have the potential as a regenerative therapy for cerebral ischemia by improving functional outcomes. However, cell transplantation has some limitations, including a low rate of the grafted cell survival. There is still a major challenge of promoting the harmonious symbiosis between grafted cells and the host. Acupuncture can effectively improve the functional outcome after cerebral ischemia. The present study evaluated the therapeutic effects and explored the mechanism of combined medial ganglionic eminence (MGE) neural progenitors differentiated from human embryonic stem cells (hESCs) with electroacupuncture (EA) in a bilateral common carotid artery occlusion (2VO) rat model. The results showed that EA could promote the survival of the grafted MGE neural progenitors differentiated from hESCs and alleviate learning and memory impairment in rats with cerebral ischemia. This may have partially resulted from inhibited expression of TNF- and IL-1 and increased vascular endothelial growth factor (VEGF) expression and blood vessel density in the hippocampus. Our findings indicated that EA could promote the survival of the grafted MGE neural progenitors and enhance transplantation therapy's efficacy by promoting angiogenesis and inhibiting inflammation.

Monitoring dynamic neurochemical signals in the brain of free-moving animals remains great challenging in biocompatibility and direct implantation capability of current electrodes. Here we created a self-supporting polymer-based flexible microelectrode (rGPF) with sufficient bending stiffness for direct brain implantation without extra devices, but demonstrating low Young's modulus with remarkable biocompatibility and minimal position shifts. Meanwhile, screening by density functional theory (DFT) calculation, we designed and synthesized specific ligands targeting Mg2+ and Ca2+, and constructed Mg-E and Ca-E sensors with high selectivity, good reversibility, and fast response time, successfully monitoring Mg2+ and Ca2+ in vivo up to 90 days.

Olfactory neuroblastoma (ONB), sinonasal undifferentiated carcinoma (SNUC), and sinonasal neuroendocrine carcinoma (SNEC) are rare malignancies arising from the sinonasal tract with limited therapeutic options. The expression of the somatostatin receptor 2 gene (), which is expressed in other neuroendocrine neoplasms and is therapeutically actionable, has been reported in these tumors. Here, we analyzed gene expression and its associations with genomic features, established biomarkers predicting of immune response, and the tumor immune microenvironment in a cohort of ONB, SNUC, and SNEC tumor samples (26, 13, and 8 samples, respectively) from a real-world database.

The Role of Parvalbumin Interneurons in Autism Spectrum Disorder.

J Neurosci Res

October 2024

Department of Central Laboratory, Shanghai Children's Hospital, School of Medicine, Shanghai Jiaotong University, Shanghai, China.

As an important subtype of GABAergic interneurons, parvalbumin (PV) interneurons play a critical role in regulating cortical circuits and neural networks. Abnormalities in the development or function of PV interneurons have been linked to autism spectrum disorder (ASD), a neurodevelopmental disorder characterized by social and language deficits. In this review, we focus on the abnormalities of PV interneurons in ASD, including quantity and function and discuss the underlying mechanisms of impairments in PV interneurons in the pathology of ASD.

Artificial Meshed Vessel-Induced Dimensional Breaking Growth of Human Brain Organoids and Multiregional Assembloids.

Brain organoids are widely used to model brain development and diseases. However, a major challenge in their application is the insufficient supply of oxygen and nutrients to the core region, restricting the size and maturation of the organoids. In order to vascularize brain organoids and enhance the nutritional supply to their core areas, two-photon polymerization (TPP) 3D printing is employed to fabricate high-resolution meshed vessels in this study.

Article Synopsis
  • Cardiac arrest can lead to significant brain injury and high mortality rates, making treatment difficult.
  • Researchers are exploring the use of neural stem cells (NSCs) for therapy, but their effectiveness is often limited after being transplanted into the body.
  • A new study shows that using a specific monosaccharide analog (TProp) enhances the function and repair capabilities of NSCs, improving recovery in animal models after brain injury due to cardiac arrest.
