The melanocortin-4 receptor (MC4R) plays an important role in appetite. Agonist ligands that stimulate the MC4R decrease appetite, while antagonist compounds increase food consumption. Herein, a functional mixture-based positional scan identified novel MC4R antagonist sequences. Mixtures comprising a library of 12,960,000 tetrapeptides were screened in the presence and absence of the NDP-MSH agonist. These results led to the synthesis of 48 individual tetrapeptides, of which 40 were screened for functional activity at the melanocortin receptors. Thirteen compounds were found to possess nanomolar antagonist potency at the MC4R, with the general tetrapeptide sequence Ac-Aromatic-Basic-Aromatic-Basic-NH. The most notable results include the identification of tetrapeptide [, Ac-DPhe(pI)-Arg-Nal(2')-Arg-NH], an equipotent MC4R antagonist to agouti-related protein [AGRP(86-132)], more potent than miniAGRP(87-120), and possessing 15-fold selectivity for the MC4R versus the MC3R. These tetrapeptides may serve as leads for novel appetite-inducing therapies to treat states of negative energy balance, such as cachexia and anorexia.

Kaolinite stands out as a promising natural geomaterial for developing new therapeutic systems aimed at addressing global health challenges, such as multidrug-resistant infections. In this study, we report on the formulation and biological activity of a therapeutic mixture composed of white micronized kaolinite (KAO) and Ziziphora essential oil (ZEO), intended for topical application on infected wounds. GC-MS analysis revealed that the primary component of ZEO is pulegone, constituting 72.

Aldehyde-functionalization of chitin nanocrystals via SI-ARGET ATRP of lignin-derived monomers.

Carbohydr Polym

Department of Polymer Materials and Engineering, College of Materials and Metallurgy, Guizhou University, Guiyang 550025, China. Electronic address:

Chitin nanocrystals (ChNCs), prepared from a down-sizing process from chitin, have recently captured great attention to access sustainable nanomaterials. The surface modification of ChNCs is crucial to regulate the surface physicochemical properties and introduce specific functions, thus satisfying their diverse applications. In this study, aldehyde-functionalized ChNCs (ChNCs-PVMA) with enhanced hydrophobicity were developed via surface-initiated activators regenerated by electron transfer for atom transfer radical polymerization (SI-ARGET ATRP) of a lignin-derived polymerizable aldehyde monomer, vanillin methacrylate (VMA).

The Baltic Sea is one of the world's most contaminated seas with long-standing adverse health status of its wildlife such as the Baltic Sea salmon, resulting in reduced fecundity and increased mortality. While adverse health effects have been reported among wild fish from the Baltic Sea, the toxicity mechanisms underlying these adversities, and the chemical effect drivers mediating them are poorly understood. To address this knowledge gap, we utilized the zebrafish (Danio rerio) embryo model to determine molecular and functional effects brought on by exposure to a technical mixture including 9 organohalogen compounds detected in serum from wild-caught Baltic Sea salmon.

Recognizing the challenges in using botanicals as sustainable pest control agents due to compositional variation, this study addresses the limitations of traditional component-based approaches such as Hewlett and Plackett or Wadley's models. Based on the assumption of noninteractivity among constituents, these models often fail to predict outcomes accurately due to dynamic intermolecular interactions. We introduce a whole mixture-based approach, employing a combination of experimental design and polynomial modeling.

Structure-Activity Relationship of Pyrrolidine Pentamine Derivatives as Inhibitors of the Aminoglycoside 6'--Acetyltransferase Type Ib.

Antibiotics (Basel)

July 2024

Center for Applied Biotechnology Studies, Department of Biological Science, College of Natural Sciences and Mathematics, California State University Fullerton, Fullerton, CA 92831, USA.

Resistance to amikacin and other major aminoglycosides is commonly due to enzymatic acetylation by the aminoglycoside 6'--acetyltransferase type I enzyme, of which type Ib [AAC(6')-Ib] is the most widespread among Gram-negative pathogens. Finding enzymatic inhibitors could be an effective way to overcome resistance and extend the useful life of amikacin. Small molecules possess multiple properties that make them attractive for drug development.
