A thy- tetracycline-resistant mutant of Streptomyces granaticolor was prepared by mutagenesis of the parental strain ETH 7437. The mutant exhibits a different morphology and an overproduction of granaticins. The ability to form an aerial mycelium and spores has been lost. The mutant cells have a round or an atypical shape and a thick cell wall, the membraneous system is enlarged by numerous mesosomes. Division septa are formed rarely. The mutant is more sensitive to both low and high temperature than the parental strain. The altered features are stably maintained for many generations. Ribosomal proteins of the mutant do not differ substantially from those of the original strain indicating that the mutant phenotype is not due to an alteration at the translational level.

Geldanamycin, a benzoquinone ansa antibiotic, has been extensively applied in medical, agricultural, and health research areas due to its antitumor, antifungal, herbicidal, and antiradiation effects. In this study, an improvement of geldanamycin production by FIM18-0592 was first performed by protoplasts combined with UV mutagenesis and ribosome engineering technology, respectively. The results showed that strains induced by UV mutagenesis of protoplasts were superior to protoplasts treated with erythromycin in terms of the positive variability, average relative titer, and maximum relative titer, with values of 51.

Identifying hormone-like quorum sensing (QS) molecules in streptomycetes is challenging due to low production levels but is essential for understanding secondary metabolite biosynthesis and morphological differentiation. This work reports the discovery of a novel γ-butenolide-type signaling molecule (SFB1) via overexpressing its biosynthetic gene (orf18) in Streptomyces fradiae. SFB1 was found to be essential for production of tylosin through dissociating the binding of its receptor TylP (a transcriptional repressor) to target genes, thus activating the expression of tylosin biosynthetic gene cluster (tyl).
