This protocol describes the application of the CRISPR-Trap from designing of the gene targeting strategy to validation of successfully edited clones that was validated on various human cell lines, among them human induced pluripotent stem cells (hiPSCs). The advantage of CRISPR-Trap over conventional approaches is the complete removal of any endogenous full-length transcript from the target gene. CRISPR-Trap is applicable for any target gene with no or little coding sequence in its first exon. Several human cell lines and different genes have so far been edited successfully with CRISPR-Trap.

Lattice sulfur-impregnated nanoscale zerovalent iron (S-nFe) has been recognized as a promising groundwater remediation agent. However, little information is available on its reactivity with ubiquitous extracellular antibiotic resistance genes (eARGs) in anaerobic groundwater, and how S content and speciation affect their interactions. Here, the efficient anaerobic degradation of eARGs by S-nFe (6 log within 5 min), resulting in completely inhibited transformation is showed.

Accidental vascular catheter removal (AVCR) by patients with cognitive impairment can result in loss of access for infusion therapy, significant blood loss, air embolism with large bore catheters, and withholding life-sustaining dialysis treatment. Chest-to-back (CTB) tunneling of central venous catheters is a technique described in the Rapid Assessment of Vascular Access Exit Site and Tunneling Options (RAVESTO) for patients at risk for AVCR. In this retrospective review, data was collected for all venous access devices inserted using CTB tunneling in patients requiring medium to long term intravenous access for infusion or hemodialysis, who had a history of self-removing medical devices due to an alteration in mental status, or patients with severely impaired skin integrity.

Porous Glass with Layered Morphology Prepared by Phase Separation.

Materials (Basel)

March 2025

State Key Laboratory of Marine Resource Utilization in South China Sea, Hainan University, Haikou 570228, China.

In this study, porous glass with controllable layered structure was successfully prepared by the phase-separation method, with the aim to develop a high-performance high-temperature catalytic (denitrification) material. Glass compositions with different R values (n (NaO)/n (BO)) were designed based on the phase diagram of sodium borosilicate glass. The layered porous structure was obtained by heat treatment in the phase-separation temperature range and acid-leaching treatment to remove the boron-rich phase.
