A Comparative Study on Egg Yolk IgY Production with Different Adjuvants and their Inhibitory Effects on . | LitMetric

AI Article Synopsis

  • Atopic dermatitis (AD) is a common skin condition in kids that can worsen with specific bacterial colonization, and a specific egg yolk antibody (IgY) has been shown to inhibit this growth.
  • The study compared the effects of different adjuvants used in the immunization of hens to produce anti-IgY antibodies, noting that oil-based Freund's incomplete adjuvant (FIA) led to the most effective antibodies.
  • Among the purified antibodies, FIA-IgY demonstrated the strongest growth inhibition of bacteria, suggesting that oil-based adjuvants are crucial for developing IgY as a potential treatment for AD symptoms.

Article Abstract

Atopic dermatitis (AD) is one of the most common skin disorders in infants and children and is often aggravated by increased () colonization. An inhibitory effect of a specific egg yolk antibody (IgY) on growth was demonstrated in this study. Furthermore, the effects of water- or oil-based adjuvants on the preparation of anti- IgY and hen immunization were compared. Hens were immunized intramuscularly with formalin-killed mixed with either a water-soluble polysaccharide -carrageenan, oil-based Freund's complete adjuvant (FCA), or Freund's incomplete adjuvant (FIA). Anti- IgYs (FIA-IgY, FCA/FIA-IgY, and Carra-IgY) were purified from the egg yolk of immunized hen eggs, and the activity of the IgY against antigen was measured by ELISA. The proportion of each IgY that was absorbed by was also determined. Then, the effect of purified anti- IgY on growth inhibition was investigated . The yolk of eggs and purified FIA-IgY from the FIA group showed the highest antibody activity, followed by FCA/FIA-IgY and Carra-IgY. The proportion of each IgY that was absorbed by antigen was as follows: FIA-IgY (18.1%), FCA/FIA-IgY (12.9%), and Carra-IgY (7.0%). Only FIA-IgY significantly inhibited growth in liquid medium. A specific IgY that was produced using the FIA adjutant inhibited growth. Although water-soluble -carrageenan showed an adjuvant effect on anti- IgY induction in egg yolk, but did not inhibit growth. The use of the oil adjuvant FIA was necessary in the preparation of anti- IgY as a treatment for AD symptoms.
