Self-assembling ferritin nanoparticle technology is a widely used vaccine development platform for enhancing the efficacy of subunit vaccines by displaying multiple antigens on nanocages. The dengue virus (DENV) envelope domain III (EDIII) protein, the most promising antigen for DENV, has been applied in vaccine development, and it is essential to evaluate the relative immunogenicity of the EDIII protein and EDIII-conjugated ferritin to show the efficiency of the ferritin delivery system compared with EDIII. In this study, we optimized the conditions for the expression of the EDIII protein in , protein purification, and refolding, and these optimization techniques were applied for the purification of EDIII ferritin nanoparticles.

Four, eight or twenty C3 symmetric protein trimers can be arranged with tetrahedral, octahedral or icosahedral point group symmetry to generate closed cage-like structures. Viruses access more complex higher triangulation number icosahedral architectures by breaking perfect point group symmetry, but nature appears not to have explored similar symmetry breaking for tetrahedral or octahedral symmetries. Here we describe a general design strategy for building higher triangulation number architectures starting from regular polyhedra through pseudosymmetrization of trimeric building blocks.

The bioinspired synthesis of virus-like silica nanoparticles in biomedical applications makes it possible to utilize the cellular delivery capabilities of viruses while minimizing the cytotoxicity of inorganic silica. In this study, we developed a diatom-inspired method for synthesizing silica-layered nanocages utilizing R5 peptide-functionalized virus-like particles (VLPs). R5 peptides were genetically inserted into the F-G loop of human papillomavirus 16 L1 proteins (HPV16 L1-R5).

The successful application of fluorescence immunoassays for clinical diagnosis requires stable photoluminescent materials and highly efficient signal amplification strategies. In this work, the magnetism-functionalized lanthanide MOF-on-MOF (FeO@SiO@MOF-on-MOF) was synthesized through intermolecular (van der Waals) interaction-assisted growth and further homogeneous epitaxial growth, which significantly improved the fluorescence performances and uncovered the underlying mechanism. The quantum chemical theory calculation and experimental studies revealed that the introduced magnetic FeO@SiO not only endowed magnetic separation capability but also promoted fluorescence performances, which increased the energy transfer of the intersystem crossing process and suppressed the luminescence of ligands and aggregation-induced quenching.

Viral infections in animals continue to pose a significant challenge, affecting livestock health, welfare, and food safety, and, in the case of zoonotic viruses, threatening global public health. The control of viral diseases currently relies on conventional approaches such as inactivated or attenuated vaccines produced via platforms with inherent limitations. Self-assembling ferritin nanocages represent a novel vaccine platform that has been utilized for several viruses, some of which are currently undergoing human clinical trials.
