Loss of pressure-induced vasoconstriction increases susceptibility to renal and cerebral vascular injury. Favored paradigms underlying initiation of the response include transient receptor potential channels coupled to G protein-coupled receptors or integrins as transducers. Degenerin channels may also mediate the response. This review addresses the ) evolutionary role of these molecules in mechanosensing, ) limitations to identifying mechanosensitive molecules, and ) paradigm shifting molecular model for a VSMC mechanosensor.

Variants of uncertain significance (VUS) represent variants that lack sufficient evidence to be confidently associated with a disease, thus posing a challenge in the interpretation of genetic testing results. Here we report an improved method for predicting the VUS of Arylsulfatase A (ARSA) gene as part of the Critical Assessment of Genome Interpretation challenge (CAGI6). Our method uses a transfer learning approach that leverages a pre-trained protein language model to predict the impact of mutations on the activity of the ARSA enzyme, whose deficiency is known to cause a rare genetic disorder, metachromatic leukodystrophy.

Background And Aims: Genome size varies by orders of magnitude across land plants, and the factors driving evolutionary increases and decreases in genome size vary across lineages. Bryophytes have the smallest genomes relative to other land plants and there is growing evidence for frequent whole genome duplication (WGD) across the lineage. Lake Tanganyika is unique among African lakes in the diversity of the pelagic top predators in the genus , where four species are endemic to the lake. Using a combination of reduced-representation and whole genome resequencing data, and pairing these with phylogenetic and demographic modeling approaches, we find that colonization of Lake Tanganyika was much more recent (∼1-2 Mya) than other major and diverse clades within the lake.
