The spike protein of SARS-CoV-2 binds to the ACE2 receptor its receptor-binding domain (RBD), with the RBD-ACE2 complex presenting an essential molecular target for vaccine development to stall the virus infection proliferation. The computational analyses at molecular, amino acid (AA), and atomic levels have been performed systematically to identify the key interacting AAs in the formation of the RBD-ACE2 complex for SARS-CoV and SARS-CoV-2 with its Alpha and Beta variants. Our study uses the molecular dynamics (MD) simulations with the molecular mechanics generalized Born surface area (MM-GBSA) method to predict the binding free energy (BFE) and to determine the actual interacting AAs, as well as two quantum chemical protocols based on the density functional theory (DFT) implementation. Based on MD results, Q, Y, Q, N, T, N, Y, F, K, Y, F, Y, and L have been identified as hotspots in SARS-CoV-2 RBD, while those in ACE2 are K, K, D, D, H, D, Q, T, Y, Y, and E. RBD with Alpha and Beta variants has slightly different interacting AAs due to N501Y mutation. Both the electrostatic and hydrophobic interactions are the main driving force to form the AA-AA binding pairs. We confirm that Q, Q, N, F, K, and F in RBD are the key residues responsible for the tight binding of SARS-CoV-2 with ACE2 compared to SARS-CoV. RBD with the Alpha variant binds with ACE2 stronger than the wild-type RBD or Beta. In the Beta variant, K417N reduces the binding, E484K slightly enhances it, and N501Y significantly increases it as in Alpha. The DFT results reveal that N, Q, Y, T, G, G, and G in RBD of SARS2 form pairs specific hydrogen bonding with Q, H, E, D, Y, Q, and K in ACE2.

Obesity is a global epidemic often associated with serious medical complications such as diabetes, hypertension and metabolic dysfunction-associated steatohepatitis. Considering the multifactorial nature of these diseases, medicinal plants could be a valuable therapeutic strategy as their phytoconstituents interact with multiple and relevant biological targets. In this context, Ilex paraguariensis emerges as a potential alternative to treat obesity and associated metabolic diseases since several studies have demonstrated its anti-inflammatory, anti-obesity and anti-diabetic effects.

The aromatic aldehyde synthase (AAS), PonAAS2, from the gall-inducing sawfly has been identified as a biosynthetic enzyme for indole-3-acetic acid (IAA), a key molecule of the plant hormone auxin, which is thought to play a role in gall induction. Unlike other insect AASs that convert Dopa, PonAAS2 uniquely converts L-tryptophan (Trp) into indole-3-acetaldehyde, a precursor of IAA. In this study, an examination of AAS enzymes from various insect species revealed that the ability to convert Trp has been acquired in only a very limited taxonomic group.

Background: This study employed melanin synthesized by Aspergillus flavus and Aspergillus carbonarius to inhibit the production of mycotoxins and bioremediation of heavy metals (HMs).
Methods: First, twenty fungal isolates were obtained from soil samples, and were evaluated to produce melanin. The melanin of the most potent producers has undergone several confirmatory experiments, including, Dihydroxyphenylalanine (DOPA)-inhibitor-kojic acid pathway detection, High-performance liquid chromatography (HPLC), Fourier-transform infrared (FTIR) and Nuclear magnetic resonance (NMR).

African Americans (AAs) with end-stage kidney disease (ESKD) experience significant barriers to accessing living donor kidney transplantation (LDKT), largely due to individual and systemic factors, including a lack of trust in healthcare systems resulting from a legacy of and continued experiences with medical racism. This cross-sectional study analyzed survey data from 416 AA patients with ESKD undergoing transplant evaluation in 2019-2023 at two kidney transplant centers in the Southeast United States, examining whether trust (specifically trust in kidney doctors, hospitals, and healthcare) modifies the relationship between attitudes towards LDKT and behavioral intentions to discuss LDKT with family and friends. Multivariable analyses revealed significant interactions.
