Identification and monitoring of cell heterogeneity from plasmid recombination during limonene production.

Appl Microbiol Biotechnol

January 2025

NUS Synthetic Biology for Clinical and Technological Innovation (SynCTI), National University of Singapore, Kent Ridge, 117456, Singapore.

Detecting alterations in plasmid structures is often performed using conventional molecular biology. However, these methods are laborious and time-consuming for studying the conditions inducing these mutations, which prevent real-time access to cell heterogeneity during bioproduction. In this work, we propose combining both flow cytometry and fluorescence-activated cell sorting, integrated with mechanistic modelling to study conditions that lead to plasmid recombination using a limonene-producing microbial system as a case study.

Functional regression for SERS spectrum transformation across diverse instruments.

January 2025

Department of Physics and Astronomy, Franklin College of Arts and Sciences, University of Georgia, Athens, Georgia 30602, USA.

Surface-enhanced Raman spectroscopy (SERS) holds remarkable potential for the rapid and portable detection of trace molecules. However, the analysis and comparison of SERS spectra are challenging due to the diverse range of instruments used for data acquisition. In this paper, a spectra instrument transformation framework based on the penalized functional regression model (SpectraFRM) is introduced for cross-instrument mapping with subsequent machine learning classification to compare transformed spectra with standard spectra.

Purpose: To evaluate Descemet membrane endothelial keratoplasty (DMEK) for failed endothelial keratoplasty (EK).

Methods: We reviewed 362 consecutive DMEK cases for failed EK (Descemet stripping endothelial keratoplasty (DSEK) or DMEK), 118 for early EK failure, and 244 for late failure in 333 eyes of 323 patients without penetrating keratoplasty. Rejection and graft survival rates were assessed with Kaplan-Meier analysis.

Nanopore Sequencing Allows Recovery of High-Quality Completely Closed Genomes of All Species from Powdered Infant Formula Overnight Enrichments.

Microorganisms

November 2024

Microbial Methods Development Branch, Division of Food and Environmental Safety, Office of Applied Microbiology and Technology (OAMT), Office of Laboratory Operations and Applied Science (OLOAS), Human Foods Program, Food & Drug Administration, College Park, MD 20740, USA.

Precision metagenomic approaches using Oxford Nanopore Technology (ONT) sequencing has been shown to allow recovery of complete genomes of foodborne bacteria from overnight enrichments of agricultural waters. This study tests the applicability of a similar approach for genome recovery from powdered infant formula (PIF) overnight enrichments, where typically dominates the overall microbiome (>90%). This study aimed to test whether using ONT sequencing of overnight PIF enrichments could recover a completely closed genome for further genomic characterization.

and are two species belonging to the Plusiinae subfamily within the Noctuidae family. Due to their morphological similarity, the identification of their larvae is difficult and time-consuming. A rapid and accurate identification of these two species is essential for their management as these species exhibit differential susceptibilities to insecticides and crops engineered to express () proteins, and a molecular tool can easily provide this differentiation.
