Hypothalamic dysfunction is an initial event following diet-induced obesity, primarily involving areas regulating energy balance such as arcuate nucleus (Arc) and median eminence (ME). To gain insights into the early hypothalamic diet-induced alterations, adult CD1 mice fed a high-fat diet (HFD) for 6 weeks were studied and compared with normo-fed controls. Transmission and scanning electron microscopy and histological staining were employed for morphological studies of the ME, while Raman spectroscopy was applied for the biochemical analysis of the Arc-ME complex. In HFD mice, ME β2-tanycytes, glial cells dedicated to blood-liquor crosstalk, exhibited remarkable ultrastructural anomalies, including altered alignment, reduced junctions, degenerating organelles, and higher content of lipid droplets, lysosomes, and autophagosomes. Degenerating tanycytes also displayed an electron transparent cytoplasm filled with numerous vesicles, and they were surrounded by dilated extracellular spaces extending up to the subependymal layer. Consistently, Raman spectroscopy analysis of the Arc-ME complex revealed higher glycogen, collagen, and lipid bands in HFD mice compared with controls, and there was also a higher band corresponding to the cyanide group in the former compared to the last. Collectively, these data show that ME β-tanycytes exhibit early structural and chemical alterations due to HFD and reveal for the first-time hypothalamic cyanide presence following high dietary lipids consumption, which is a novel aspect with potential implications in the field of obesity.

This article studies the synthesis, as well as the structural, vibrational, and optical properties of Eu-doped ZnO quantum dots (QDs) and investigates the energy transfer mechanism from the ZnO host to Eu ions using Reisfeld's approximation. Eu-doped ZnO QDs at varying concentrations (0-7%) were successfully prepared using a wet chemical method. The successful doping of Eu ions into the ZnO host lattice, as well as the composition and valence states of the elements present in the sample, were confirmed through X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS) analyses.

We have grown (111)- and (001)-oriented NiO thin films on (0001)-Sapphire and (001)-MgO substrates using pulsed laser deposition (PLD), respectively. DC magnetic susceptibility measurements underline that the Néel temperatures of the samples are beyond room-temperature. This is further confirmed by the presence of two-magnon Raman scattering modes in these films in ambient conditions.

A highly sensitive aptamer sensor (aptasensor) is proposed based on metal-organic frameworks-silver nanoparticles (AgNPs@MOF) to detect sulfadimethoxine (SDM) by surface-enhanced Raman spectroscopy (SERS). AgNPs@MOF with SERS activity was successfully fabricated by synthesizing AgNPs in situ on the surface of MIL-101(Fe), and SDM aptamer and Raman reporter 4-aminophenthiophenol (4-ATP) were selected as specific recognition elements and signal probes, respectively. When SDM was absent, the SDM aptamers were effectively adsorbed on the surface of AgNPs@MOF, thus keeping AgNPs@MOF in a dispersed state, resulting in a weakened SERS signal of 4-ATP.
