The backbone extraction process is pivotal in expediting analysis and enhancing visualization in network applications. This study systematically compares seven influential statistical hypothesis-testing backbone edge filtering methods (Disparity Filter (DF), Polya Urn Filter (PF), Marginal Likelihood Filter (MLF), Noise Corrected (NC), Enhanced Configuration Model Filter (ECM), Global Statistical Significance Filter (GloSS), and Locally Adaptive Network Sparsification Filter (LANS)) across diverse networks. A similarity analysis reveals that backbones extracted with the ECM and DF filters exhibit minimal overlap with backbones derived from their alternatives.

Background: Women are disproportionately affected by Alzheimer's disease (AD) and exhibit greater AD neuropathology than men. Women possess two X chromosomes, with one randomly silenced across each cell for dosage compensation. X chromosome inactivation (XCI) is not complete, and XCI-escaping genes provide a promising avenue of discovery for biological pathways driving sex-specific AD risk.

Background: Published data have highlighted associations between Alzheimer's disease (AD) susceptibility loci and AD-related brain changes. The amyloid imaging to prevent AD (AMYPAD) consortium is a European collaboration consisting of several parent cohorts, four of which had raw genotype array data available. We sought to integrate and harmonise the genetic data, calculate AD polygenic risk scores (PRS), and investigate their association with global amyloid deposition.

Background: Two-thirds of Alzheimer's Disease (AD) cases are women, and our team has identified molecular factors that relate to disease in a sex-specific manner. Here, we leverage single-cell transcriptomics from dorsolateral prefrontal cortex (N = 424) from the Religious Orders Study and Memory and Aging Project (ROS/MAP; AD Knowledge Portal syn2580853) to characterize sex-specific contributors at cellular resolution.
Method: Single-nucleic RNAseq data was generated and processed as previously described.
