Tissue clearing combined with high-resolution confocal imaging is a cutting-edge approach for dissecting the three-dimensional (3D) architecture of tissues and deciphering cellular spatial interactions under physiological and pathological conditions. Deciphering the spatial interaction of leptin receptor-expressing (LepR) stromal cells with other compartments in the bone marrow is crucial for a deeper understanding of the stem cell niche and the skeletal tissue. In this study, we introduce an optimized protocol for the 3D analysis of skeletal tissues, enabling the visualization of hematopoietic and stromal cells, especially LepR stromal cells, within optically cleared bone hemisections.

This study extends the Job Demands-Resources (JD-R) framework by proposing JD-R 3.0, which explores the dynamic and nonlinear relationships among Work Connectivity Behavior After-Hours (WCBA), Perceived Organizational Support (POS), and employee outcomes, with a specific focus on psychological contract violation (PCV) and psychological distress. Data were collected from 625 employees in a large financial organization in China across two waves, and polynomial regression and response surface analysis were used for hypothesis testing.

Continuum solvation models such as the polarizable continuum model and the conductor-like screening model are widely used in quantum chemistry, but their application to large biosystems is hampered by their computational cost. Here, we report the parametrization of the Miertus-Scrocco-Tomasi (MST) model for the prediction of hydration free energies of neutral and ionic molecules based on the domain decomposition formulation of COSMO (ddCOSMO), which allows a drastic reduction of the computational cost by several orders of magnitude. We also introduce several novelties in MST, like a new definition of atom types based on hybridization and an automatic setup of the cavity for charged regions.

This study presents a comprehensive workflow for developing and deploying Multi-Layer Perceptron (MLP)-based soft sensors on embedded FPGAs, addressing diverse deployment objectives. The proposed workflow extends our prior research by introducing greater model adaptability. It supports various configurations-spanning layer counts, neuron counts, and quantization bitwidths-to accommodate the constraints and capabilities of different FPGA platforms.
