A New Highly Deuterated [F]AV-45, [F]D15FSP, for Imaging β-Amyloid Plaques in the Brain.

[F]AV-45 (florbetapir f18, Amyvid) is an FDA-approved PET imaging agent targeting Aβ plaques in the brain for diagnosis of Alzheimer's disease (AD). Its metabolites led to a high background in the brain and large bone uptake of [F]F, produced from dealkylation of the PEG chain. To slow down the in vivo metabolism, we report the design, synthesis, and evaluation of a highly deuterated derivative, [F]D15FSP, and compared it with -methyl-deuterated [F]D3FSP and nondeuterated [F]AV-45. D15FSP displayed excellent binding affinity ( = 7.52 nM) to Aβ aggregates. In vitro autoradiography of [F]D15FSP, [F]D3FSP, and [F]AV-45 showed excellent binding to Aβ plaques in human AD brain sections. Biodistribution studies displayed lower bone uptake at 120 min for [F]D15FSP compared to that for [F]D3FSP and [F]AV-45 (1.44 vs 4.23 and 4.03%ID/g, respectively). As the highly deuterated [F]D15FSP displayed excellent Aβ binding affinity, high initial brain penetration, and lower bone retention, it might be suitable for PET imaging in detecting Aβ plaques.

Hydrogen-atom transfer is crucial in a myriad of chemical and biological processes, yet the accurate and efficient description of hydrogen-atom transfer reactions and kinetic isotope effects remains challenging due to significant quantum effects on hydrogenic motion, especially tunneling and zero-point energy. In this paper, we combine transition state theory (TST) with the recently developed constrained nuclear-electronic orbital (CNEO) theory to propose a new transition state theory denoted CNEO-TST. We use CNEO-TST with CNEO density functional theory (CNEO-DFT) to predict reaction rate constants for two prototypical gas-phase hydrogen-atom transfer reactions and their deuterated isotopologic reactions.

A Study on the Photoisomerization of ()-Dehydrozingerone, Its ()-()-C₂ Symmetric Dimer, and Their -Methylated Derivatives.

In this study, UV-induced ()-to-() geometrical isomerizations of the curcumin degradation product ()-dehydrozingerone, along with curcumin-inspired ()--methylated dehydrozingerone and their corresponding C-symmetric dimers, were investigated. All compounds produced corresponding () isomers in varying yields upon UV irradiation in deuterated solvents. The efficiency of these photoisomerizations depended on the solvent and wavelength used.

Mechanistic Studies on the Gold-Catalyzed Intramolecular Hydroalkylation of Ynamides to Indenes.

An in-depth experimental and computational study to rationalize the mechanism underlying the gold-catalyzed intramolecular hydroalkylation of ynamides to indenes is reported. Evaluating the reactivity of a set of deuterated ynamides and gold complexes allowed to get valuable insights into the mechanism of this reaction, while DFT calculations allowed to determine a plausible reaction pathway for this unprecedented transformation. This pathway involves the activation of the ynamide followed by a [1,5]-hydride shift from the highly reactive, in situ generated keteniminium ion, and a subsequent cyclization before deprotonation followed by a final protodeauration.
