Recent Advances in Photo-mediated Radiofluorination.

Carbon-fluorine bond formations have received a lot of attention because organofluorine compounds are widely used in pharmaceutical, agricultural, and materials science applications. In particular, the incorporation of fluorine-18, which is a commonly used radioisotope for radiopharmaceuticals for positron emission tomography (PET), a molecular imaging tool for the visualization of biochemical events, human metabolism processes, and the measurement and diagnosis of diseases in humans, plays a crucial role in clinical and preclinical studies. Several synthetic methodologies for carbon-fluorine-18 bond formation have been developed. However, conventional fluorination methods have some remaining drawbacks such as the high temperature and basic environment. Photo-induced catalysis is an emerging technique that allow chemists to achieve the synthesis of target molecular architectures under mild conditions. Moreover, several radiofluorination strategies have been developed via photocatalysis. In this review, we focused on describing recent advances in the field of light-mediated radiofluorination.

Photomediated reversible addition fragmentation chain transfer (RAFT) step-growth polymerization is performed using a trithiocarbonate-based chain transfer agent (CTA) and acrylate-based monomers both with and without a photocatalyst. The versatility of photo-mediated RAFT step-growth is demonstrated by one-pot synthesis of a graft copolymer via sequential monomer addition. Furthermore, oxygen-tolerant photo-mediated RAFT step-growth is demonstrated, facilitated by the appropriate selection of photocatalyst and solvent pair (zinc tetraphenyl porphyrin [ZnTPP] and dimethyl sulfoxide [DMSO]), enabling ultralow volume polymerization under open-air conditions.

Liquid-liquid phase separation (LLPS) is responsible for the emergence of intracellular membrane-less organelles and the development of coacervate protocells. Benefitting from the advantages of simplicity, precision, programmability, and noninvasiveness, light has become an effective tool to regulate the assembly dynamics of LLPS, and mediate various biochemical processes associated with LLPS. In this review, recent advances in optically controlling membrane-less organelles within living organisms are summarized, thereby modulating a series of biological processes including irreversible protein aggregation pathologies, transcription activation, metabolic flux, genomic rearrangements, and enzymatic reactions.

Metal-free organic photocatalysts for photo-mediated reversible deactivation radical polymerization (photo-RDRP) are witnessed to make increasing advancement in the precise synthesis of polymers. However, challenges still exist in the development of high-efficiency and environmentally sustainable carbon dots (CDs)-based organocatalysts. This review discusses recent advances in light-driven radiochemistry for three key isotopes: fluorine-18, carbon-11, and zirconium-89, and their applications in positron emission tomography (PET). In the case of fluorine-18, the predominant approach involves the use of cyclotron-produced [F]fluoride or reagents derived thereof. Light serves to activate either the substrate or the fluorine-18 labeled reagent.
