Easy and Efficient Permeabilization of Cyanobacteria for Enzyme Assays Using B-PER.

Cyanobacteria are photosynthetic bacteria that thrive in diverse ecosystems and play major roles in the global carbon cycle. The abilities of cyanobacteria to fix atmospheric CO and to allocate the fixed carbons to chemicals and biofuels have attracted growing attentions as sustainable microbial cell factories. A better understanding of activities of enzymes involved in the central carbon metabolism might lead to increased product yields. Currently, cell-free lysates are widely used for the determination of intracellular enzyme activities. However, due to thick cell walls in cyanobacteria, lysis of cyanobacterial cells is inefficient and often laborious. The present protocol describes an easy and efficient method to permeabilize cyanobacterial cells, without lysing them, and direct usage of the permeabilized cells for the determination of metabolic enzyme activities .

To accelerate the water dissociation in the Volmer step and alleviate the destruction of bubbles to the physical structure of catalysts during the alkaline hydrogen evolution, an integrated electrode of cobalt oxide and cobalt-molybdenum oxide grown on Ni foam, named CoO-Co2Mo3O8, is designed. This integrated electrode enhances the catalyst-substrate interaction confirmed by a micro-indentation tester, and thus hinders the destruction of the physical structure of catalysts caused by bubbles. Electrochemical testing shows the occurrence of a surface reconstruction of the integrated electrode, and CoO is transformed into Co(OH)2, denoted as Co(OH)2-Co2Mo3O8.

A green approach towards the synthesis of both conventional and magnetic fluorescent powders for revealing latent fingerprints (FPs) is disclosed. The powders formulation is based on a biodegradable matrix and fluorescent dyes extracted from commercial felt-tip markers. Two classes of powders are described: one with a fluorescent component, and other with both fluorescent and magnetic components.
