A ready-to-use hetero-tricyclic compound, 5,5'-(furan-2,5-diyl) bis (1,3,4- oxadiazol-2-amine) (5), was synthesized with a good yield; it has an suitable fluorescence characteristic and research founded that it can respond to trace Pd in water at a normal pH range. A fluorescence titration revealed the detection limit for Pd was as low as 3.97 × 10 M. Density-functional theory calculation using Guassian09 implied that the breakage of conjugation and coplanarity of compound 5 led to fluorescence quenching. Compound 5 could be applied as a chemical probe to detect trace amounts of Pd with good accuracy, fast response time, excellent selectivity, and high sensitivity. FT-IR, NMR, and MS were used to characterize the chemical structure of compound 5.

Endogenous tagging enables the study of proteins within their native regulatory context, typically using CRISPR to insert tag sequences directly into the gene sequence. Here, we introduce qTAG, a collection of repair cassettes that makes endogenous tagging more accessible. The cassettes support N- and C-terminal tagging with commonly used selectable markers and feature restriction sites for easy modification.

Multiplexed immunofluorescence (IF) can be achieved using different commercially available platforms, often making use of conjugated antibodies detected in iterative cycles. A growing portfolio of pre-conjugated antibodies is offered by the providers, as well as the possibility for conjugation. For many conjugation methods and kits, there are limitations in which antibodies can be used, and conjugation results are sometimes irreproducible.

Dye-encoded bead-based assays are widely used for diagnostics. Multiple bead populations are required for multiplexing and can be produced using different dye colors, labeling levels, or combinations of dye ratios. Ready-to-use multiplex bead populations restrict users to specific targets, are costly, or require specialized instrumentation.

The reliable application of field deployable fluorescent dissolved organic matter (fDOM) probes is hindered by several influencing factors which need to be compensated. This manuscript describes the corrections of temperature, pH, turbidity and inner filter effect on fluorescence signal of a commercial fDOM probe (fDOM). For this, Australian waters with wide ranging qualities were selected, e.
