We describe the synthesis, photophysical properties, and photodynamic activity of a methemoglobin (metHb) wrapped covalently by human serum albumins (HSAs) incorporating protoporphyrin IX (PPIX): a metHb-HSA -PPIX cluster. The metHb core catalyzes H O disproportionation to generate O in tumor tissue. The HSA -PPIX shell acts as a photosensitizer for O formation. The metHb-HSA -PPIX cluster acts as a dual functional protein drug for photodynamic therapy.

The autofluorescence of erythrocyte porphyrins has emerged as a potential method for multi-cancer early detection (MCED). With this method's dependence on research-grade spectrofluorometers, significant improvements in instrumentation are necessary to translate its potential into clinical practice, as with any promising medical technology.
