Multicomponent protein complexes called inflammasomes play a major role in the innate immune system by activating proinflammatory cytokines and promoting a highly inflammatory form of programmed cell death, called pyroptosis. A hallmark of the function of the nucleotide-binding domain, leucine-rich repeat and NLRP3-mediated inflammasome assembly is the polymerization of ASC into large filaments. The ASC filaments recruit and activate procaspase-1 by induced proximity. We developed an assay for monitoring the polymerization of the pyrin domain of ASC by microscale thermophoresis. We have validated the assay by analyzing the effects of buffer conditions, mutations of ASC and the use of seeds on the polymerization behavior of ASC.

Cutaneous colonization with Staphylococcus aureus (SA) is frequently observed in patients with atopic dermatitis. SA produces a wide variety of bacterial toxins, among which δ-toxin was found to induce degranulation of mast cells. Degranulation of mast cells could enhance bacterial clearance and protection from future SA infection but lead to exacerbation of atopic dermatitis.

Background: Alcoholic Liver Disease (ALD) is a hepatic disorder resulting from prolonged or excessive alcohol intake. The predominant manifestation of ALD is fatty liver, which progresses to alcoholic hepatitis as the disease worsens. Pyroptosis is a novel type of programmed cell death that is intricately linked to the inflammatory cascade, presenting a promising avenue for therapeutic intervention in the management of ALD.

Background: Microplastics (PS-MPs) are a new type of pollutant with definite hepatotoxicity. Selenium, on the other hand, has natural, protective effects on the liver.

Objectives/methods: The purpose of this experiment is to find out whether nano-selenium (SeNP) can alleviate liver damage caused by microplastics.

Atopic dermatitis (AD) is a prevalent skin disorder worldwide. However, many AD medications are unsuitable for long-term use due to low therapeutic efficacy and side effects. Extracellular vesicles (EVs) extracted from mucus have demonstrated therapeutic efficacy in AD.

Article Synopsis
  • - The flower of Gomphrena globosa (G. globosa) is recognized in traditional Chinese medicine for its potential liver-protective properties, particularly through its polysaccharide components that may combat fatty liver and hepatitis.
  • - Researchers isolated a specific polysaccharide called GGL0.05S1 from G. globosa, which was found to inhibit fat accumulation and oxidative stress in liver cells, showing effectiveness in preventing liver damage in animal models.
  • - The mechanism behind GGL0.05S1's protective effects involves enhancing the antioxidant capacity of the thioredoxin protein, promoting fat oxidation and cellular cleanup processes, while also reducing inflammation through the inhibition of the NLRP3 signaling pathway.
